Objective:Under the guidance of the traditionary raditional chinese medicine theory,using the technology of metabonomics and the breakthrough point of the whole metabolites in disease as well as biomarkers,to study fever syndro-me induced by2,4-dinitrophenol in rat and therapy effect of Scutellariae Rad-ix-Phellodendri Chinensis Cortex and Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus,the Chinese drugs for treatment of fever syndrome.Method:By UPLC-TOF/MS technology platform,to analyze the vitro composition and Constituents absorbed into blood of Scutellariae Radix-Phellodendri Chinensis Cortex and Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus,and copy the fever syndrome animal model as the carrier,with the bitter cold of the compound traditional Chinese medicine intervention,their urine samples will be tested to draw assistance from UPLC-TOF/MS and to deal information with MassLynx V4.1and changes in biomarkers to evaluate the intervention effect,to investigate the efficacy of traditional Chinese medicine herbs and relevance.Results:1.Using UPLC-TOF/MS to analyze and to distinguish the vitro composit-ion of Scutellariae Radix-Phellodendri Chinensis Cortex’s and Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus’s aqueous extract.20piece chemical composition are distinguished in Scutellaria baicalensis Georgi-Cortex Phellodendri Chinensis and31piece in Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus.2. Combining the analysis results of vitro component,by ESI+mode, the Scutellariae Radix-Phellodendri Chinensis Cortex Group found29components into the rat blood,5of them were original in form, and23others were metabolites;34components were detected in the rat blood with Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus,which prototype components were6,24metabolites.By ESI-mode,Scutellariae Radix-Phellodendri Chinensis Cortex Group found3prototype components a nd10metabolites;Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeni ae Fructus found4prototype components and13metabolites。3.Copy of2,4-dinitrophenol-induced fever syndrome animal model to macrosigns and metabolomics approach to evaluation model,the model successfully reproduced;Make sure the fever syndrome can be characterized by positive ion mode of46potential biomarkers,and identified two of them the structure of biological markers,these two biomarkers for biological analysis,respectively,and tyrosine,tryptophan metabolism.4.Using Scutellariae Radix-Phellodendri Chinensis Cortex and Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus intervention by fever syndrome model to the macro signs and metabolomics approach to evaluation.Two groups, respectively21、24a potential biomarker trends with a callback,show that the join of Gardenia,enable the skullcap cork intervention enhanced role.Conclusions:1.In analysis and identification of the vitro composition results are consistent with the main active components of Scutellaria、Phellodendron、 Gardenia drugs.Joined the Gardenia,the types of active ingredients and the increase in the number may greatly improve the therapeutic effect of the purging fire detoxification.2.Results can be seen from the blood components of Scutellariae Radix-Phellodendri Chinensis Cortex and Scutellariae Radix-Phe llodendri Chinensis Cortex-Gardeniae Fructus.Fewer prototype components were detected in the two treatment groups,mostly in the form of metabolites.Detected by protoype components,respectively baicalin,wogonosi de,berberine and geniposide,these four components were representative chemical composition of Scutellaria、Phellodendron、Gardenia.3.Based on the Huanglianjiedu decoction,compatibility in the form of the two study(Scutellariae Radix-Phellodendri Chinensis Cortex and Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fructus), to copy the DNP fever syndrome animal model as the carrier, the metabolomics technology and UPLC-TOF/MS analysis of means,from the Global and the metabolites of biological markers in support of Scutellariae Radix-Phellodendri Chinensis C ortex and Scutellariae Radix-Phellodendri Chinensis Cortex-Gardeniae Fruct-us.
