| In this thesis, fluorescence properties of phellodendrine and papaverine, active ingredients of traditional Chinese medicine, are studied. A new method, three dimensional fluorescence spectra coupled with second-order calibration(3DF-2OC) method, for determination of phellodendrine in medicinal materials Phellodendri Amurensis Cortex(Guanhuangbo in Chinese) and Phellodendri Chinensis Cortex(Huangbo in Chinese), is established. The feasibility of using 3DF-2OC method for determination of papaverine in Papaveris Pericarpium(Yingsuqiao in Chinese) medicinal materials is investigated, and the content of papaverine in Yingsuqiao sample is determined by a HPLC method.Three parts are included in the thesis:1. Fluorescence spectra of phellodendrine and papaverine are studied. In acidic aqueous solution, phellodendrine can produce strong fluorescence, with excitation wavelengths λex 230,280 nm, and emission wavelength λem 315 nm. Using quinine sulfate as a reference,fluorescence quantum yield of phellodendrine is measured to be 0.30. The fluorescence of papaverine aqueous solution is very weak. In strong alkaline condition, addition of β-CD into papaverine solution can enhance the fluorescence, λex/λem = 293/359 nm, fluorescence quantum yield is 0.016.2. Based on the fluorescence property of phellodendrine, a 3DF-2OC method for determination of phellodendrine in Guanhuangbo and Huangbo medicinal materials is proposed. Contents of phellodendrine in Guanhuangbo reference crude herb and in commercial crude herb are determined to be 0.149% and 0.232%, respectively. Contents of phellodendrine in Huangbo referenee crude herb and in commercial crude herb are determined to be 0.425% and 0.336%, respectively. These results are verified by a HPLC method, indicating that the proposed method is reliable.3. Based on the sensitized fluorescence property of papaverine, a 3DF-2OC method for determination of papaverine in Yingsuqiao medicinal materials is investigated, and failed to get satisfactory results. Using a HPLC method, contents of papaverine in Yingsuqiao reference crude herb and in commercial crude herb are determined to be 0.028% and 0.035%,respectively. The recovery rate, precision and stability of the method are all right. |
