Air-dried roots of Ixeris sonchifolia (Bge.) Hance were extracted with EtOH at roomtemperature. After removal of EtOH, the water suspension was re-extracted with petroleumether, ethyl acetate and n-BuOH respectively.11compounds were separated from ethyl acetatelayers using the column chromatography, HPLC and other separation methods. Thestructures of these compounds were confirmed based on their physicolchemical properties andthe spectrum technology,especially using the NMR spectrum. The compounds extracted fromthis plant are3-hydroxy-1(10),3-oxoguaia-eno-12,6-lactone-2-ketone (1), SonchifoliasolidB (2),3-hydroxy-11-H-1(10),3-oxoguaia-eno-12,6-lactone-ketone (3),3-hydroxy-1(10),11(13),3-oxoguaia-trieno-12,6-lactone-2-ketone (4), caffeic acid (5),3,4-dihydroxy-benzoic acid(6),15-hydroxy-1(10),11(13),3-oxoguaia-trieno-12,6-lactone-2-ketone (7), Sonchifoliasolid D (8), Sonchifoliasolid A (9),4,8-(3’-methoxy-4’-hydroxy)-diphenyl-ditetrahydrofuranxylogen(10),(+)-Syringaresinol (11).While among these, the compounds5,6,10and11are extracted from this plant for the firsttime.Sonchifoliasolid A is the essential component of the Ixeris sonchifolia. Here we usingthe two different methods(boiling with water and extracted with EtOH) to discuss thedifferent results of extracting Sonchifoliasolid A from the root of Ixeris sonchifolia and the itshows that the method using EtOH to isolated Sonchifoliasolid A from this plant not only hasa higher yield but also easier compared with water boiling.So exrtaction with EtOH is the bestmethod to isolation Sonchifoliasolid A from the root of Ixeris sonchifolia.In order to get more Sonchifoliasolid A aglycone to proceed the structure modification ofthese compounds, we using many methods like acidolysis, enzymolysis and so on to try to getthe it with high yield and the enzymolysis with b-glucolase shows the bestresult(yield:94.7%).Here the relationship between the structure of the sesquiterpene lactones compoundsexracted from the Ixeris sonchifolia and activity were discussed. Results of the antitumoractivity of several compounds show that Sonchifoliasolid A has no activity while theaglycone has a good activity of antitumor. To this guaiane style sesquiterpene lactonescompounds, the effect of the2-position to the activity is very small,while the12and6position have obvious effection on the antitumor activity and the activity will reduced if thering opened. The9-position has a certain extend influence to the activity of the compoundmaybe related to the hydrophile-hydrophobic balance of the substituent group.
