Objective: Lead poisoning anemia animal models were established, expel lead effectand improve blood system of APS were observed. The effect of angelica polysaccharide(APS) on expelling lead and improving blood system was investigated.Method:21day-old SD rats were selected and established lead poisoning model, Lead-induced anemia by ip Lead acetate. normal group(0.9%NaCl), model group, APS15,30,60,120mg/kg groups, negative control group(DMSA). The whole blood lead levels in rats weredetected by graphite furnace atomic absorption spectrometry, Hb content in rats weredetected by GaoTieFa cyanide, RBC count in rats were tested by adopting the microscopecount method. Hct determination in rats were detected by blood analyzer, The whole bloodALAD in rats were detected by paradimethylamino-benzaldehyde method colorimetric, Theoriginal ZPP in rats were detected by zinc porphyrin blood fluorescence detector. The levelof serum IL-6, EPO were measured by ELISA method. Data statistical analysis andchart-making were made by SPSS13.0and SigmaPlot10.0.Results:①All APS dose treatment groups of anemia effect and lead time of administration istime-effect relations. To medicine,0day,7day14days each APS BPb level as time dosegroup of increased significantly reduced, Hb, RBC content increases with the increase of thetime(P <0.01).②The effect of APS and DMSA on BPb level of SD rats: Compared with normalcontrol group, BPb level was significantly increased in other groups(P<0.01); blood leadlevel in each intervention group significantly lower than model control group(P<0.01);Compared with DMSA group, there were no statistically significan.(P>0.05) ③The effect of APS and DMSA on Blood parameters of SD rats: compared withnormal group, Hb, RBC and Hct in model control group were significantly decreased(P<0.01); compared with model control group, the levels of Hb, RBC and Hct in eachintervention group significantly increased (P<0.01). However, there were no significantdifference among model control group and APS groups (P>0.05).④The effect of APS and DMSA on ALAD activity of SD rats: ALAD activity weresignificantly decreased in model control group than normal control group(P<0.01); ALADactivity in each APS group and DMSA group was significantly increased than the modelcontrol group(P<0.01); Compared with DMSA group there were no statisticallysignifican(P>0.05).⑤The effect of APS and DMSA on ZPP of SD rats: ZPP were significantly increasedin model control group than normal control group(P<0.01); ZPP in each intervention groupsignificantly decreased than the model control group(P<0.01), Compared with DMSA groupthere were no statistically significan(P>0.05).⑥The effect of APS and DMSA on serum IL-6, EPO of SD rats: The levels of serumIL-6、EPO were significantly decreased in moder cntrol group than normal controlgroup(P<0.01); The levels of serum IL-6、EPO were significantly increased in each APSgroup than moder control group(P<0.01);15~60mg/kg APS were does-effect relations,60mg/kg APS was the best tincrease of serum IL-6, EPO; Compared with DMSA group,EPO was significantly increased in APS60mg/kg group(P<0.05); There is no effect ofDMSA on serum IL-6(P>0.05).Conclusion:1、APS has the obvious effects on expelling lead and improving blood system,2、The protective effects of APS on the blood system by promoting secretionof serum EPO, IL-6.
