Objective:The topic is based on the Neural immune inflammation hypothesis and the hypothesis of β-amyloid protein cascades, utilizing HE stain and IHC to examine the neuronal morphological structure of the AD rats’Hippocampus and the variations of the NF-KB and IκB-α protein expression, to ascertain the effects of waternut herb extract on AD rats caused by Aβ1-40on cellular and molecular level and preliminarily discuss its mechanism of action.Method:The topic is on the basis of the hypothesis of P-amyloid protein cascades, using Stereotaxic instrument to determine the skull projective position of the unilateral hippocampal CA1sector of the rats and quantitatively inject Aβ1-40aggregation fragments to obtain AD animal models. Observing the morphological and structural changes of the injected hemispherical Hippocampus cells of the rats by HE stain, together with the variations of the NF-KB, IκB-α and caspase-3protein content in the AD rats’ Hippocampus by IHC, preliminarily ascertain the effects of waternut herb extract on AD rats caused by Aβ1-40, and provide the experimental basis for the further mechanism research.Results:1.The structure of the AD rats’Hippocampus in Model Group is abnormal; the cell zone is in disorder and diffused distribution; the neuronal loss is obvious and the cell body shrinks, with pyknosis and spongiocyte hyperplasia. The NF-KB protein expression increases and the IκB-α protein expression decreases; the caspase-3protein expression increases too; there is an obvious distinction compared with the Normal Group.2.Compared with the Model Group, the arrangement of the cell structure of the AD rats’Hippocampus in Western Medicine Group is relatively ordered; the neurons increase, and are in normal shape and homogeneous distribution. The NF-KB protein expression decreases and the IκB-α protein expression increases; the caspase-3protein expression decreases too; there is an obvious distinction.3.Compared with the Model Group, the cell structure and neurons of the AD rats’Hippocampus in Treatment Group have improved in different degrees. The NF-KB, IκB-α and caspase-3protein expression also has the same trend of change, which is: Alcohol-high Group> Water-high Group> Alcohol-low Group> Water-low Group.Conclusions:1.The waternut herb extract can inhibit the schizolysis of the NF-KB and IκB-α by increasing the content of IκB-α in the AD rats’ Hippocampus, avoiding the genetic transcription caused by the NF-KB entering the nucleolus, thereby inhibit the activation of caspase-3and avoid the apoptosis, protecting the normal morphology and function of the neurons.2.By analyzing the experimental data, the effect of the Alcohol-high Group waternut herb extract exceeds that of the identical amount of the Water-high Group waternut herb extract, therefore presumed that the active ingredient for healing AD in waternut herb is primarily found in80%alcoholic extract.
