Objective Establish the rats model of obstructive jaundice,to study the change of intestinal epithelial paneth cell function in obstructive jaundice rats and its relationship with intestinal mucosal barrier.Methods Choose 27 healthy adult male SD rats randomly divided into three groups,A blank Control group (the Control group C, n = 9), Control group B (Sham operation,SO group, n = 9), Obstructive jaundice group C (Obstructive jaundice, OJ group, n =9). Group A is blank control group without any treatment; Group B as control, open only separation the common bile duct; As the experimental group, line double ligation bravery manager. Three groups of rats were put to death after bile duct ligation laparotomy observation in the 9th day,then, return the corresponding specimens. To return the inferior vena cava blood clinical laboratory detection of ALT, AST, TBIL,DBIL liver function index; To return for portal vein blood for DAO and D-lactic acid ELISA detection; Return 10% formaldehyde fixed terminal ileum application,reserved for HE staining and immunohistochemical.Results 1 .Animal model: (1) animal model was successfully established.Macroscopic view tube ligation, 1 day which is a mild jaundice, to the skin, especially for the ears, with the extension of time in progression, urine yellow, 2 days 3 days rats skin yellow dye, decreased activity, continue to have urine yellow performance with defecate color white, postoperative rats obviously limited overall activities,consciousness is poorer, had mild sleepiness.2. The liver function: SO group and Control group in serum ALT, AST, TBIL, DBIL levels have no significant difference;OJ group rats serum ALT, AST, TBIL, DBIL reflect the function of liver and biliary obstruction of indexes such as content increased significantly, compared with other two groups have significant difference(P<0.01).3. Intestinal mucosa: (1) intestinal mucosa histological observation: SO group and the Control group rats ileum mucosa villus structural integrity, intestinal mucosa columnar epithelial cells .OJ group intestinal villus back structure is incomplete,the number and density decreased, clearance between mucosal villus increased,accompanied by some hair loss and cell swelling, mucosal glands arranged relatively sparse, goblet cells reduce obviously, mucosal atrophy thinning, rendering with interstitial hyperemia edema and nonspecific inflammatory cells infiltrating inflammatory change.(2) bowel mucosal villus height and crypt depth: SO group compared with the Control group of rats, intestinal mucosal villus height and crypt depth had no significant difference, the villus height and crypt depth of OJ group was obviously lower than and shallow in the other two groups (P < 0.01).4. ELISA: D - lactic acid and the content of the DAO: SO group and Control group in serum lactic acid and DAO D - content has no obvious difference;OJ group content of serum lactic acid and DAO D - less than the marked increase in the other two groups(P<0.01).5. Immunohistochemical of SIgA: SIgA positive product mainly locate in the lamina propria of the cell. the Control group and SO group more visible sIgA protein expression, and there was no significant difference;OJ group sIgA protein expression quantity compared with other two group significantly reduced (P < 0.05).6. The observation of intestinal Paneth cell: SO group and the Control group rats by lined up at the bottom of the small intestinal Paneth cell,a cone-shaped, irregular nucleus, near the cell base, a massive amount of cytoplasm topped with eosin dye red round secretory granules.OJ group rats by reduced Numbers of cells, secretory granules dyeing becomes shallow.7. Immunohistochemical of Paneth cell: lysozyme positive product mainly located in the small intestine fossae by cell cytoplasm, compared with OJ group, SO group and Control group shows more lysozyme positive cells expression.Calculation of intestinal mucosa lysozyme positive cell percentage accounted for, compared with the Control group and SO group, OJ group of lysozyme protein expression of positive cells was significantly reduced (P < 0.01).8. Correlation analysis: lysozyme as a kind of antibacterial proteins secreted by cells,in the expression of OJ group and negatively correlated with D - the content of lactic acid (r = -0.95, P < 0.01), and the expression of sIgA were positively correlated (r =0.89, P< 0.01).Conclusion obstructive jaundice can cause liver damage and intestinal mucosal barrier dysfunction.At the same time, the obstructive jaundice can cause by reduced Numbers of cells and function disorder.Correlation analysis showed that under the condition of obstructive jaundice by cell changes associated with a significant function of intestinal mucosa.
