Objective:Through the literature to review the different cultivation and processing methods of Coptis chinensis Franch., and the herbs content by HPLC, establish the fingerprint characteristics. To prefer chuan huanglian cultivation technology and processing methods, to lay the foundation for chuan huanglian environmental biotechnology research and ecpand production.Method:By the method of consulting ancient of Chinese material and chorography, to analyz the historical records of Coptis chinensis Franch., including the distribution of germplasm resources, cultivation techniques and processing methods. Through field trips and visits to, compare the cultivation techniques and processing characteristics of different habitats Sichuan huanglian. Using of botany, medicine identification and plant anatomy, to compara different habitats medicinal herbs and anatomical. Combining natural medicine chemistry, instrumental analysis and traditional Chinese medicine chemistry, using HPLC determines the content of Jatrorrhizine, Epiberberine, Coptisine, Palmatine and Berberine. Using Chinese medicine chromatographic fingerprint evaluation system bulids stable Huanglian chemical fingerprint feature; use SPSS19.0to cluster analysis the differences of different origin Sichuan Huanglian.Result:Through the research of materia medica ancient,"shujun" and "wuyang kawadani" were the main producing areas of ancient huanglian, consistenting with western Sichuan producing areas and east Sichuan producing areas now. The results of fieldwork and literature analysis showed that the ground temperature and light intensity were one of the key factors in the impact of the quality of herbs. In the origin processing technology, in situ semi-dry in western Sichuan was the most cost saving(reducing supplies and transportation); the circular pit pool was the most reasonable(heating uniformity). The different medicine traits comparison results showed that the trait differences with the original processing method. HPLC with basic mobile phase gradient elution can make the chromatographic perk of Jatrorrhizine, Epiberberine, Coptisine, Palmatine and Berberine separately, stably and reproducly. Acidic condtions cannot sepatate the Epiberberine and Jatrorrhizine, but the acidic and alkaline mobile phase to establish the two chromatographic fingerprint can obtain12common peaks, similarity≥0.99. Qualitative and quantitative cluster analysis results show that huanglian is not with the origin, but related to the cultivar.Conclusion:Western Sichuan and east Sichuan are the road producing areas of chuan huanglian, with excellent quality. The berberine alkaloid content order of Coptis by different cultivation methods:the traditional natural shed> natural battlefield shed> natural understory> awning> wild forest. To promote in situ semi-dry the circular pit pool, to reduce processing costs and ensure the quality of medicine. HPLC with the basic mobile phase can be used as one of the methods to control the huanglian quality, and to determine the content. The clustering results show that to pay attention to variety selection and seed cultivation in the chuan huanglian cultivation and production.
