Objective: To testify the existence of " marker of cold and hot substance " and toprovide the modern scientific evidence for the property of TCM drugs and porper clinicalusage of it, the Effects of Coptis Chinensis Franch and it’s CCC substance and CCHsubstance in Rats with Excessive Heat Syndrome on body temperature, independentactivities, chills and fever trend, ATP enzyme activity, Energy metabolism, substancemetabolism, thyroid hormone levels and cytokine activity were observed. The resultswere analysed with multivariate analysis of variance and PLS model, and the functioncharacteristics and pharmacodynamic mechanism were estimated.Methods: Wistar rats were randomly divided into blank control group, modelcontrol group, Coptis Chinensis Franch group, the CCC group and the CCH group, Thereare five rats in every groups.The syndrome model group,the CCC group and the CCHgroup were induced by subcutaneous injection of2,4-dinitrophenol.And the bank controlgroup were induced by normal saline.The change of the body temperature andindependent activities, warm and cold preferring tendence were observed after treatedwith Coptis Chinensis Franch and it’s extract of CCC and CCH. Chemical colorimetryspectrophotometry was used to detect pyruvic acid, lactic acid, hepatic glycogen,triglyceride and succinate dehydrogenase, lactate dehydrogenase, ATP enzyme activity.Automatic microplate method was used to detect the content of rat plasma T3, T4, TSH,TRH, IL-1β, IL-6, IL-8, TNF-α, the results were estimated by the PLS regression model and multivariate analysis of variance method.Results: Coptis Chinensis Franch and it’s extract of CCC and CCH could reducesignificantly the body temperature,independent activities, warm and cold preferringtendence, ATP enzyme activity and hepatic energy charge in rats with excessive heatsyndrome. These extracts could also inhibit the function of hypothalamus-pituitary-thyroid axis and energy metabolism, substance metabolism, reduce thyroxine secretion,inhibit cytokine activity. The effects in Coptis Chinensis Franch is best. The effects inCCC group is better than that in CCH group with PLS method. These results coincidencewith HPLC-DAD fingerprint analysis. It proved the objective existence of " marker ofcold and hot substance "Conclusions: Coptis Chinensis Franch and it’s extract of CCC and CCH could inhibit materialmetabolism, energy metabolism, thyroid hormone and cytokine activity in rats with excessive heatsyndrome. it may testify to some extent the objective existence of " marker of cold and hot substance", which provided a certain reference for the research and clinical applications to heat syndromes ofcoptis chinensis franch.
