Qingwen Baidu prescription derived from the warm febrile diseases monograph Yi Zhen YiDe, which was written by Yu Shi-yu in1794in the Qing Dynasty, and was included in the people’sRepublic of China Veterinary Pharmacopoeia of2005and2010edition. Qingwen Baiduprescription was made up with14traditional Chinese medicines, they are Plaster, Rehmannia,Cornu bubali, Coptidis rhizome, Gardenia augusta Merr., Cortex moutan, Scutellaria, Paeoniaeradix, Figwort root, Anemarrhenae, Forsythia suspense, Platycodon root, Glycyrrhiza uralensisand Lophatherum herb, it has been used in clinical for more than two hundred years, and hassignificant therapeutic effect to heat hair spot and high fever coma.. Our object is to changedosage form by modern means on the foundation of pharmacopoeia prescription, in order to fitwith the modern medication principles and keep the intrinsical therapeutic effect. There must bequality control standards in accordance with the dosage form to ensure security and controllability.Therefore, we systematically studied the preparation technology and quality criteria, evaluated thestability of products, effects of toxicology and pharmacology.Objective: To ascertain preparation technology of Qingwen Baidu Keli, to study the qualitymonitoring, and to investigate its stability. Evaluting toxicity of Qingwen Baidu Keli.We alsoinvestigated influences of Qingwen Baidu Keli to mice immune organs to evaluate its immuneenhancement effects to mice.Methods: Optimization of extraction technologies, which contains extraction technology ofberberine hydrochloride, baicalin and other medicines in the complex prescription. Ascertainforming technology by optimization of adjuvant and wetting agents. We established qualityevaluation system by methodology investigation. To control the quality, we adopted platechromatography, and made quality differentiation for Coptidis rhizome, Forsythia suspense,Gardenia augusta Merr. and Anemarrhenae in Qingwen Baidu Keli. The stability of products wasalso determined according to regulations in the people’s Republic of China VeterinaryPharmacopoeia; we also evaluated toxicity of Qingwen Baidu Keli through acute toxicity test andsubchronic toxicity test;effects of Qingwen Baidu Keli to immune organs of mice was used toevaluate its immune enhancement effects.Results: Coptidis rhizome needs decoction three times using10,8and8times water for1.5h,1.5h and1h, respectively; Scutellaria also needs to decoction three times using10,8and8timeswater for2h,1h and1h, respectively, then combined the fluid, filtered and condensed.Hydrochloric acid solution of2mol/L was used to adjust pH to1.0-2.0at80℃, kept thetemperature for1h, and standing for12h. The sedimentation was added with6-8times water, and 40%NaOH was used to adjust the pH to7.0, the partes aequales ethanol was added, agitated tohaemolyze, and filtered again, hydrochloric acid solution of2mol/L was used to adjust pH to2.0at60℃, kept the temperature for30min, and standing for12h. The sedimentation was washedwith alcohol until pH was7.0, the sedimentation can be used once alcohol was exhausted. The rest12medicines, first digeratur for1h in water, then decoction for twice,2hours for the first time and1hour for the second time, filtered, and combine the filtrate. Combined all thick paste andscutellaria powder, then quantity sufficient adjuvant were added to prepare granula.Plate chromatography was used to quality differentiation of Coptidis rhizome, Forsythiasuspense, Gardenia augusta Merr. and Anemarrhenae in Qingwen Baidu Keli. The results showedthat the preparation technology was reasonable and feasible, quality evaluation system wasconsummate and reliability, through the observation experiments of remaining samples, threebatches of samples in plastic conditions was studied, and the stability is well in six months.Toxicologic study:Qingwen Baidu Keli is low toxicity through acute toxicity test,and it issafety and reliability in the clinical. Qingwen Baidu Keli has no influence to grouth of rats throughsubchronic toxicity test, it is also non-toxic reaction when repeate to administrate Qingwen BaiduKeli for42days.Qingwen Baidu Keli has certain immune enhancement effection.Conclusion: Qingwen Baidu Keli has good stability. Low toxicity.The therapeutic effect ofenhancement of immunity is evident. Its successful development can provide a new formulationfor veterinary clinical usage of classical formula.
