ObjectiveThe8-hydroxy-2-Deoxyguanosine (8-OHdG) was produced by theDNA(deoxirybonucleic acid) oxidation. Recently, the relationship between8-OHdG andthe development of gastric cancer not fully understood. In our study, we examined theserum and tissue expression of8-OHdG in patients with gastric cancer and normal patients.This study was aimed to determine the role8-OHdG played in the development of gastriccancer.Patients and Methods30patients with gastric cancer,15patients with chronic gastritis,20healthy controlswere included in our present study. The level of serum8-OHdG was determined byenzyme-linked immunosorbent assay. The level of tissue8-OHdG was measured byimmunohistochemisty (IHC).ResultsThe level of serum8-OHdG in patients with gastric cancer (23.8±13.2pg/ml) washigher than that in the healthy controls (9.9±5.0pg/ml). The difference between them wassignificant (p<0.05).The tissue8-OHdG expression in patients with gastric cancer was significantlyincreased than dysplasia patients and healthy controls (p<0.05).The expression of8-OHdG in gastric cancer was positively related to thedifferentiated degrees of the tissues (p<0.05), which means more8-OHdG were detected inpoorly intermediately differentiated adenocarcinoma than in those highly differentiatedones; the expression of8-OHdG in gastric cancer was obviously related to the lymphaticmetastasis, depth of invasion and clinical stages (p<0.05). It indicated that the8-OHdGlevel is higher in the lymphatic metastasis(p<0.05),penetrated serosa (p<0.05), and PhaseIII-IV(p<0.05) than in those without lymphatic metastasis,without invasion and Phase I-II,respectively. However, the expression of8-OHdG in gastric cancer did not relate to the patient’s age and sex (p>0.05).ConclusionThe level of serum8-OHdG and tissue expression of8-OHdG were significantlyincreased in gastric cancer patients than dysplasia patients and healthy controls. Thissuggested that the change of8-OHdG expression may played an very important role in thedevelopment of gastric cancer.8-OHdG expression are positively correlated to differentiated degrees of the tissues,the depth of invasion, lymphatic metastasis and clinical stages, meanwhile, it has nothingto do with the patient’s age, sex. This implies the upregulating levels of8-OHdG issignificantly correlated with some malignant behaviours of gastric cancer.
