Astilbe chinensis (Maxim.) Franch. et Sav belongs to Astilbe chinensis and has been used for chronic bronchitis and arthralgia in traditional Chinese medicine.It mainly distributes to China态Russia态Japan and so on. The bergenin of Astilbe chinensis (Maxim.) Franch. et Sav has been collect<China Pharmacopoeia (2010Edition)> and has been used for arthralgia. The study was based on the research of further investigate to the technology that the bergenin has been extracted the Astilbe chinensis (Maxim.) Franch.et Sav, and the best harvest time, and else to investigate the Antioxidant activity and effect of bergenin in inhibition of human colon cancer SW1116cell.A comprehensive review was finished based on overall literature related to resource, chemical constituents and pharmacological activity of the plants of the Astilbe chinensis (Maxim.) Franch.et Sav, which provided useful information for its further study on chemical constituents and pharmacological activity.We studied on the technology, the content of bergenin from the different extractions and different solvent of Astilbe chinensis (Maxim.) Franch.et Sav was determined by HPLC.To optimize the extraction process for bergenin by central composite design and response surface methodology怂Analysis indicated the optimum conditions of bergenin were64%ethanol85minutes for once reflux,17fold solvent and three times for extraction. It shows that the optimum process is simple and more convenient,with an higher precision.According to the method of following, the extract of Astilbe chinensis (Maxim.) Franch.et Sav was partitioned between petroleum ether,acetic ether and n-butanol. The parts of acetic ether were repeatedly subjected to the silica gel column chromatography, respectively, and3compounds were isolated from them, in which3compounds were identified as bergenin, daucosterol,Ī²-sitosterol by MS,1H-NMR,13C-NMR.The content of the bergenin from the different harvest periods of Astilbe chinensis (Maxim.) Franch.et Sav determined by Alltech HPLC. After in the mid of September, the content of bergenin to the accumulation of saturated gradually in various collecting periods,and Stems and leaves are not contain the bergenin.The content of flavonoids and polysaccharides from the different harvest periods and different parts of Astilbe chinensis (Maxim.) Franch.et Sav were determined by UV spectrophotometry. The contents of total flavonoids from Astilbe chinensis (Maxim.) Franch.et Sav decreased in July, other months are not significant difference. The contents of total polysaccharides from Astilbe chinensis (Maxim.) Franch.et Sav are higher in June and October,and lower in May and August than others. The highest contents of total polysaccharides in October up to23.79%.Different parts of Astilbe chinensis(Maxim.)Franch.et Sav contains flavonoids and polysaccharides. The sequence of it is root> stem> leaf.In this study, determination different polar parts in the root of Astilbe chinensis (Maxim.) Franch.et Sav in different concentrations of the antioxidant capacity怂In the method of ABTS, the ethyl acetate fraction has strong antioxidant activity.When the concentration of0.62mg/mL, the scavenging rate is99.40%. In the method of DPPH, the butanol fraction has strong antioxidant activity. When the concentration of0.62mg/mL,the scavenging rate is98.34%,it is stronger antioxidant activity than Vc.In the method of reducing power, the ethyl acetate fraction has strong reducing power. When the concentration of0.62mg/mL, the absorbance is2.541. In the determination of the phenolics content, the phenolics content of the ethyl acetate fraction is140.62(Ī¼g/20mg)To investigate the inhibitory effects of the different concentrations of bergenin on proliferation inhibition by MTT assay. The different concentrations of bergenin inhibited SW1116cells growth in an obvious dose-dependent manner.Distinct points:1态The central composite design and response surface method to optimize the formulation of the bergenin from Astilbe chinensis (Maxim.) Franch.et Sav of extraction process.2态The study confirm different polar parts in the root of Astilbe chinensis (Maxim.) Franch.et Sav in different concentrations of the antioxidant capacity:The different polar parts in the root of Astilbe chinensis (Maxim.) Franch.et Sav has strong scavenging power to ABTS+Ā·and DPPH, and it shows strong reducing power. Results showed a good dose-concentration relationship.3态Determine the optimum harvest time of Astilbe chinensis (Maxim.) Franch.et Sav:the harvest time is in the mid of September.
