Study on the establishment of experimental chronic osteomyelitisin rabbit tibiaObjectives: This study was performed to analyze the establishment of experimentalchronic osteomyelitis in rabbit tibia.Methods: Ten healthy rabbits were used in this study.0.5ml5%sodium morrhuateand0.2ml/kg5.0x106CFU/ml Staphylococcus aureus suspensions were injected intothe bone defect in inner side of right tibia. The bone defect was sealed with bone wax toprevent bacterial leakage into the surrounding soft tissues and no antibiotics forprevention of bacteremia. To observe the normal state, anus temperature and visualstudy of lesion area during the normal raising after operation.The experimentalstaphylococcus aureus osteomyelitis in rabbits was evaluated by radiologic andbacteriologic at the3th week after infection.Results:①One rabbits died in the3th day after operation, the others in poor mentalhealth showed loss of weight and reduce appetite,the temperature of rabbits rised in the1st-4th day(40.3℃-41.4℃)and recovered to normal gradually(38.5℃-39.5℃),then theykept normal in the following days. Swelling and fevering could be found from the3rdday in the surgical region which emerged soft tissue swelling,refusing touch,purulentsecretiong and sinus tract gradually.②X-ray showed soft tissue swelling in the middletibia where emerged bone cortex density decreasing, osteolytic bony destruction,periosteal proliferation and mottled osteoclasia in the outboard.③Staphylococcusaureus could be found from surgical region.Conclusion: The establishment of experimental chronic osteomyelitis in rabbit tibia canbe accomplished by injection of Staphylococcus aureus suspensions into the bone defectin inner side of right tibia. Sudy on the effect and mechanism of Tuodushengsui treating OsteomyelitisObjective: To observe the effect of Tuodushengsui on treating the rabbit models ofchronic osteomyelitis,and to study the mechanism.Methods:60rabbits animal were randomly divided into four groups: blank contrastgroup were breed normally,the other3groups were made for animal models of chronicosteomyelitis. The Tuodushengsui group were lavaged(5ml,bid) and washout(5ml,bidor qid) with the extracting solution of Tuodushengsui at the same time, Gentamicingroup and model group were washout respectively with dilution ofGentamicin(10ml,bid or qid)and physiological saline(10ml,bid or qid), All animals weretreated for2months, then observe the numeration of leukocyte srumlysozymeactivityand bone X-ray of the models on the20th,40th,60th, after the beginning of treatment.After treatment,making tissue section to get the pathological evaluation.Results:All observative time the numeration of leukocyte, srumlysozymeactivity ofmodel group were decreased by blank group(P<0.01), there were maniples recoverywith Gentamicin group but lower than blank group remainly,Tuodushengsui groupraised up than the others,60days after treating,the numeration of leukocytesrumlysozymeactivity both approached blank group(P>0.05).At the same time,diagnosis type of X-ray with Tuodushengsuifang group were better than the others. Themyeloid reaction of Tudushengsui group recessed, backbone remodeled when boneabscess and high-density shadow of sequestrum appeared on the model group, that wasthe40th days. Lots of neutrophil,lymphocyte plasmacyte and osteoclast can be foundthe model group, neutrephil and lymphocyte infiltration appeared the gentamicingroup, the Tuodushengsui group displayed tiny inflammatory and osteoblast.Conclusion: Great significant effect were found among the group ofTuodushengsui,Tuodushengsui can control the inflammation of bone well and werehelpful to bone structure recovery and modularly fasciculus recanalisation. Improvingthe immune function and repairing of bone may be the mechanism.
