| Objective:To observe the effects of Liuweidihuang Decoction to Unilatera ureteral obstruction (UUO)rats by monocyte chemotactic protein(MCP-1) and transforming growth factor(TGF-(β1), investigated the mechanism of renal Tulointerstitial fibrosis.Methods.With Sprapue-Dewley rat models UUO operation, this search oberserved the effects of Liuweidihuang Decoction on Pathologic signs of Tubulointerstitial and the expression of the MCP-1 and TGF-βin the renal tubulointerstitial.Let compare Liuweidihuang Decoction group with sham-surgery group,model group and EnalaPril group.Results:1.We observed that obstructive kidney have lesion of interstitial fibrosis manifested by pathological changes such as inflammatory cell infiltration, cell proliferation, tubular ectasia ect.3days after UUO operation, many quantitative pathological changes happened 1 week after UUO operation, research model of renal interstitial fibrosis were created successfully. 2. Observation on Pathological histomorphology of renal interstitial fibrosis caused by UUOoperation shows that there was no significant abnormality in sham operation group with HE staining, there were renal tubular epithelial cellular edema, tubular ectasia, tubal wall attenuation, interstitial hyperplasia and fibrosis in model group,and changes were more significant at the 14 th,21th day. Above-mentioned changes in Liuweidihuang Decoction group and EnalaPril group were lighter than those in model group at each time spot. Masson staining quantitative analysis shows that difference of interstitial area density in model compared with that in sham operation group at each time spot has statistical significance(p<0.05);difference of interstitial area density in Liuweidihuang Decoction group, EnalaPril group compared with that in model group at each time spot has statistical significance(p<0.05);difference of interstitial area density in liuwei Dihuang Decoction group compared with that in EnalaPril group at each time spot has statistical significance (p<0.05).3. Expression and quantitative analysis of MCP-1ã€TGF-β1 in the interstitium of kidney tubules with Immunohistochemisty shows that Expression of MCP-1ã€TGF-β1 in sham operation group was weak. MCP-1ã€TGF-β1 in model group had stronger expression compared with that in sham operation group at each time spot, mainly in stromal cells and renal tubular epithelial cell, positive expression was weak in kidney glomerulus, the difference has statistical significance. Expression of MCP-1ã€TGF-β1 in Liuweidihuang Decoction group, EnalaPril group was weak compared with that in model group at each time spot and the difference had statistical significance. Expression of MCP-1ã€TGF-βin Liuweidihuang Decoction group was stronger compared with that in EnalaPril group at each time spot and the difference has statistical significance (p<0.05).Conclusion:Liuweidihuang Decoction degraded the expression of MCP-1 and TGF-β1 in tubulointerstitial and alleviated tubulointerstitial pathology of UUO rats. It can be adjuvant drug on obstructive nephropathy. |
