| Clinical study:Clinical study on treatment of laxative constipation with the method of warning kidney to strength spleenReferring to the "Guideline for Clinical Trails of New Patent Chinese Medicines", a designed test method of randomized positive drug paralleled with clinical study is applied. The diagnostic criteria referred the current Rome III standard,30patients with laxative constipation (weak spleen and kidney) were treated with the method of warming kidney to strength spleen, meanwhile,30cases were arranged as a contract group. Fuzi formula granules:log; Shengbaizhu formula granules:30g; Roucongrong formula granules:15g; Shenghuangqi formula granules:20g; Heshouwu formula granules:15g; Danggui formula granules:log;Huomaren formulagranules:15g;Heizhimaformula granules:15g;Yizhiren formula granules:15g; Baishao formula granules:15g and Gancao formula granules:log. Oral, twice daily,1bag each time after meals. For the contrast group:Mosapride (Shandong Lunan Beit Pharmaceutical), take orally,50mg per time,3times daily, take before meals. The study results showed that there are no significant difference in age, gender, disease history and clinical symptoms before treatment in the cases selected in the two groups, after treatment, compare the treatment effects of the treatment group and the contract group after4weeks of treatment, through non-parameter inspection, Z=-4.4908,P<.0001<0.05, the difference has statistical significance, The overall efficiency of the treatment group is90%, significantly higher than50%of the contract group, this means that the treatment effects in the treatment group are superior to that of the contract group. Improvement conditions of various Chinese medical syndrome scores of the treatment group and the contract group are compared, the treatment group is significantly superior to the contract group in aspects of defecation interval, degree of difficulty in defecation, defecation time, stool shapes, etc, the difference has statistical meaning (P<0.05). There is not significant difference in treatment effects of the method of warming kidney to strengthen spleen on patients with different ages, genders, disease history and disease situations(P>0.05), this means that this treatment method has significant effects on all patients, regardless their ages, genders, disease history and disease conditions. It is confirmed by study that Chinese medicines integrated in a ways of warning kidney to strength spleen, lubricating intestine to promote defecation can significantly improve the clinical symptoms of patients with laxative constipation (weak spleen and kidney), the overall efficiency is up to90%, this is an effective treatment method.Experimental study:Study on impact of warning kidney to strength spleen on colon c-kit, SCF protein expression of rats with constipation.The animal model of rat constipation is successfully copied by using laxatives, through observing the rat feces grains, weight and shape, we can found that feces of rat in the model group are dry, the number is less and the weight is light. Through observing the intestinal transport functions, an activated carbon suspension marching method is applied, the test results show that the intestine of the rats in the model groups transports is slower, compared with the contract group, there is a significant difference.After modeling, implement treatment through the Chinese medicinal formulation developed by the method of warning kidney to strengthen spleen, observe the treatment group after a treatment course is finished, compare various indicators of the3groups of animals in the model group and the normal group, including fecal grains, weight and shapes; check the intestinal transport function:rat colon c-kit, SCF protein expression. The results show that it is an effective treatment method to treat rat with constipation with the method of warming kidney to strength spleen, including checking fecal grains, weight and shapes improvement and intestinal transport functions. Compared with the model groups, significant difference may be noticed.The study also checked the colon c-kit and SCF protein expression of experimental rat by using the Western-blot method, the results show that the normal group expression degree is higher, expression of the model group is significantly lower than the normal group, the difference had statistical significance, after treatment, the c-kit and SCF protein expression of each group is improved, for c-kit protein expression, compared the treatment group with the model group, the difference has statistical significance, there is a significant increasing in the middle dose of traditional Chinese medicine group and in the small dose of traditional Chinese medicine group and the western medicine group, also there is a significant difference between different treatment groups. For SCF protein expression, all treatment groups are significantly improved, higher than the model group, yet this has no statistical meaning, this is possible associated with less sample numbers.The study shows that the mechanism of laxative constipation rats is possibly associated with decline of c-kit and SCF protein expression, the method of warming kidney to strengthen spleen is able to improve c-kit and SCF protein expression, yet the mechanism of action is still not very clear. |
