Expression And Significance Of MiR-196b And HoxB8mRNA In Human Colorectal Cancer

Objective：To research the expression of miR-196b and HoxB8mRNA in colorectalcancer and their correlation with clinicopathologic features, to explore the relationshipbetween miR-196b and HoxB8in vivo.Methods：Expression of RNA (including miR-196b and HoxB8mRNA) and HoxB8protein were detected respectively by quantitative real-time reverse transcriptase PCRand western blot in30cases of colorectal carcinoma and corresponding normalmucous membrane tissue.Results：In colorectal carcinoma, expression of miR-196b and HoxB8mRNA werehigher than that of the control(P＜0.05). Expression of miR196b was associated withlymph node metastasis，neoplasm stages(I+II,III+IV)and distant metastasis (P＜0.05).On the other hand, no significant differences were observed regarding tumor site, size,gross type, depth of invasion, tissue differentiation, age and sex(P＞0.05). Expressionof HoxB8mRNA was no significant differences concerning lymph node metastasis,tumor stages(I+II,III+IV), distant metastasis, tumor site, size, gross type, depth ofinvasion, tissue differentiation, age and sex(P＞0.05). In colorectal cancer, expressionof HoxB8protein was lower than that of the control(P＜0.001). The expression ofmiR-196b mRNA was negatively correlated with HoxB8mRNA expression（r=-0.458，P＜0.05), and HoxB8protein expression with no obvious correlationin colorectal cancer （r=-0.236，P＞0.05).Conclusions：(1) The expressions of miR-196b and HoxB8mRNA in colorectalcancer are higher, the high expression of miR-196b mRNA is related to thetumorigenesis and progression of colorectal cancer as well as correlated withprognosis in colorectal cancer.(2) The miR-196b inhibits expression of HoxB8mRNA by binding to the3′-UTR of target HoxB8mRNA.
