| Objective:To explore the correlation of PPARαL162Vã€C2528Gã€V227Agene polyphism to early-onset T2DM and Atherosclerosis.Methods: A total of800(418men and382women)T2DM subjects wererecruited and divided into two groups:early-onset and late-onset groups which weredefined as diagnosed at≤40and≥40years of age, respectively. Early-onset T2DMsubjects400(211men and189women). Late-onset T2DM subjects400(207menand193women). Extracted peripheral blood white blood cell genomic DNA to detectPPARαL162Vã€C2528Gã€V227A polymorphism by the application of polymerasechain reaction(PCR)and calculate gene type and allele frequency of each group aswell as clinical biochemical indicatorsã€incidence of atherosclerosis.Results:1.The fasting plasma glucose(FPG), triglyceride(TG), total cholesterol(TC)are higher and insulin resistanceã€inflammatory response more severe in early-onsetT2DM group than in late-onset T2DM grop.2.The overall frequencies of LL, LV, VV in early-onset T2DM group and inlate-onset T2DM group were91.5ï¼…,0ï¼…,8.5ï¼…, and97ï¼…,0ï¼…,3ï¼…respectively.The L162allele frequency is91.5ï¼…, and V162allele frequency is8.5ï¼…inearly-onset T2DM group, the L162allele frequency is97ï¼…, and V162allelefrequency is3ï¼…in late-onset T2DM group. There was statistical differencein the gene type frequency distribution between the two groups by check-up.4.The overall frequencies of VV, VA, AA in early-onset T2DM group and inlate-onset T2DM group were91.5ï¼…,8.5ï¼…,0ï¼…, and95ï¼…,5ï¼…,0ï¼…respectively.The V227allele frequency is95.7ï¼…, and A227allele frequency is4.3ï¼…inearly-onset T2DM group, the V227allele frequency is97.5ï¼…, and A227allelefrequency is2.5ï¼…in late-onset T2DM group. There was statistical difference in thegene type frequency distribution between the two groups by check-up.5.The early-onset T2DM group V162allele has higher FPG,2hPG, HbA1C, TC,TG, hs-CRP, HOMA-IR than the late-onset T2DM group L162allele. The early-onsetT2DM group C2528allele has higher TG than the late-onset T2DM group G2528allele. The early-onset T2DM group A227allele has higher HOMA-IR than thelate-onset T2DM group V227allele.6.The early-onset T2DM group V162allele has higher incidence ofatherosclerosis than the early-onset T2DM group L162allele〠the late-onset T2DMgroup L162allele and V162allele. The early-onset T2DM group C2528allele hashigher incidence of atherosclerosis than the early-onset T2DM group G2528alleleã€the late-onset T2DM group G2528allele and C2528allele. The early-onset T2DMgroup A227allele has higher incidence of atherosclerosis than the early-onset T2DMgroup V227alleleã€the late-onset T2DM group V227allele and A227allele. Therewas statistical difference in incidence of atherosclerosis between the early-onsetT2DM group V162alleleã€C2528alleleã€A227allele and other alleles by check-up.Conclusion:1.The early-onset T2DM group V162allele has poor blood glucose control, moredyslipidemia, more severe insulin resistance and inflammatory response than thelate-onset T2DM group L162allele.2. In the early-onset T2DM group PPARαL162Vã€C2528Gã€V227A allelesfrequencies are more elevatory than the late-onset T2DM group.3.The early-onset T2DM group V162allele has poor blood glucose control,more dyslipidemia, more severe insulin resistance and inflammatory response than thelate-onset T2DM group L162allele. The early-onset T2DM group C2528allele hasmore dyslipidemia than the late-onset T2DM group G2528allele. The early-onsetT2DM group V227allele has more severe insulin resistance than the late-onset T2DM group A227allele.4. PPARαL162Vã€C2528Gã€V227A polyphism may have associations to theearly-onset T2DM and Atherosclerosis. |
