Objective:This paper on the basis of chronic obstructive pulmonary disease (COPD), TCMpathogenesis of Qi deficiency and blood stasis in airway remodeling, Select theyiqigubiao representative Fang Yupingfeng San, combined with promoting bloodcirculation drugs SanQi, on airway remodeling of COPD rat model of regulation to dopreliminary research, Through the observation of Yuping Feng San, SanQi, YupingFeng San plus SanQi effects on cytokine concentration in serum of COPD rat modelof TGF-β1, MMP-9, TIMP1, VEGF, To investigate the possible mechanism ofSupplementing Qi and activating blood circulation method in prevention andtreatment of airway remodeling in COPD, to provide ideas and methods of preventionand treatment for clinical application of new.Methods:1.To establish the rat model of COPD:75clean grade male SD rats, were randomlydivided into normal control group, COPD model of natural recovering group, YupingFeng San group, Yuping Feng San plus SanQi group, SanQi groups, Except thenormal control group, other groups were respectively in experiment first,14days tothe rat intratracheal instillation of lipopolysaccharide in200μ L; Second to28days(Fourteenth days except)for Honghe Cigarette smoking,30min/day, a total of28days; Normal control group not to smoke, but by the same method of intratrachealinstillation of normal saline. 2. Study of Yuping Feng San, SanQi interventions: intragastric administration ofexperimental animal, The normal control group and COPD model group: givennormal saline (1ml/200g body weight, every day), a total of30days; Yuping FengSan group, SanQi group, Yuping Feng San plus SanQi groups according to the clinicalequivalent dose conversion (1ml/200g body weight, every day), a total of30days.3.To observe rat behavior change; the lung tissue of rats for pathological sections, HEstaining, To observe the pathological changes; Using double antibody sandwichELISA assay for detection of TGF-β1MMP-9, TIMP1, VEGF in rat serumconcentration.Results:1. behavior changesAfter modeling, COPD model group with different degree of restlessness,breathing, With the modeling time, hair gradually lost luster, looked tired, activity wassignificantly reduced or fell down motionless, and gathered into a mass movement,nod, shortness of breath, abdominal muscle twitching, wheezing, and sneezing andoronasal secretions increase wait for a symptom, normal control group had noabnormal changes, suggests successful model.2lung tissue pathologyThe lung tissue of rats left, HE staining: Bronchial epithelium of rats in modelgroup fall off, the tube wall and surrounded by a large number of monocytes andlymphocytes, goblet cells; alveolar cavity expanding significantly, alveolar wallfracture, fusion of pulmonary alveoli, obvious emphysema; fibroblast proliferation,collagen fibers, small airway collagen fiber hyperplasia, thickening, smooth musclecells arranged in a random, smooth muscle layer thickening, lumen narrowing;capillary hyperplasia, vascular stenosis, vascular wall of the glass change. The normalgroup rats bronchial and alveolar structure is clear, the collagen increased obviously, the vessel lumen without obvious stenosis, suggests successful model.3. Effect of experimental drugs on serum COPD in rats of TGF-β1, MMP-9, TIMP-1,VEGF concentrationYuping Feng San can lower the serum of COPD rat model of TGF-β1, VEGFlevels, compared with the model group, the difference was statistically significant (P <0.05), MMP-9, TIMP-1of Yuping Feng San has a downward trend, but the differencecompared with the model group had no statistical significance (P>0.05). YupingFeng San plus SanQi, SanQi can lower the serum of COPD rat model of TGF-β1,MMP-9, TIMP-1, VEGF levels, compared with the model group, the difference wasstatistically significant (P <0.01, P <0.05); Yuping Feng San plus SanQi candown-regulate the VEGF level in serum of COPD rat model, compared with YupingFeng San,SanQi, statistically significant differences (P <0.01).Conclusion:1. By supplementing qi and activating blood circulation intervention airwayremodeling in COPD rats, reduce the bronchial and vascular visible rat lung tissuearound the infiltration of inflammatory cells, extracellular matrix deposition andcollagen fiber hyperplasia to degradation, airway wall, lumen of blood vesselinflammation can be improved.2. Yuping feng San plus SanQi is one of the mechanism of on airway remodeling ofCOPD rat model by adjusting the level of VEGF, inhibit fibroblast and endothelialcell growth, degradation of collagen synthesis and bronchial wall and vascular wallmatrix deposition, thereby relieving airway remodeling of COPD.3. Traditional Chinese medicine Yiqihuoxue prescription has some effect on thedevelopment of airway remodeling by COPD disease.
