Objective:Lipopolysaccharide COPD airway remodeling in rats by smoke plus infusion to observe SiWu Tang drugs on (Chuan Xiong-Angelica and Shudi-white peony) root decoction on rats lung tissue of pathology and related facors:TGF-β1, VEGF, MMP-9,TIMP-1, sIgA and SC concentration explore SiWu Tang drugs on(Chuan Xiong-Angelica and Shudi-white peony) on reducing COPD airway remodeling mechanism of action, Chinese medicine diagnosis and treatment of COPD airway further weight plastic provide a theoretical basis.Methods:1. Copy the animal model:smoked plus infusion by LPS method, copy the COPD airway remodeling pathological model, shared copy 28 days.2. Grouping and administration:SPF grade Wistar male rats were randomly divided into normal control group, model group, positive drug group, Siwutang, Chuanxiong -angelica,Shudi -white peony Low peony high-dose group, a total of 12 groups, each group 10. Model control group and normal saline group, positive drug group roxithromycin gavage; Chinese medicine treatment of different groups different doses of medicine gavage for 28 days.3. Experimental observation indicators:observed before and after modeling, after administration of rat lung tissue pathological changes; from rat lung tissue were measured by ELISA rat TGF-β1, VEGF, MMP-9, TIMP-1, sIgA, SC’s concentration.Results:1.COPD behavioral changes in rats:rats before and COPD consistent replication process, the intervention group were seen with varying degrees of sneezing, listlessness, decreased activity, dull hair, nodded breathing, eating less, nose and mouth secretion was increased, the body curled up and other symptoms.2.COPD rats morphological changes:drug intervention group compared with the model rats, chronic airway inflammation and bronchial mucosa cells at all levels (eg: lymphocytes, monocytes and plasma cells) and some neutrophils reduce cell; a significant reduction in the thickness of the bronchial smooth muscle tissue; goblet epithelial cells overexpressing decrease; at the same time, small pulmonary artery wall thickness is reduced.3. Changes related factor concentration COPD rat cells:①model group and the normal group, rats TGF-β1, VEGF, TIMP-1, sIgA and SC levels were increased, MMP-9 and MMP-9/TIMP-1 ratios were lower, there was a significant difference (P <0.01);② Chinese medicine treatment group compared with model group, rats TGF-β1, VEGF, TIMP-1, sIgA and SC levels were reduced, MMP-9 and MMP-9/ TIMP-1 ratio were increased, there was a significant difference(P<0.01); Siwutang,Chuanxiong-angelica,Shudi-white peony root in each dose group dose intervention group compared with the model group, its VEGF, TIMP-1, sIgA, SC detection index downregulated (P<0.01).Conclusions:1. Siwutang and drugs on (Chuanxiong -angelica,Shudi-white peony) can reduce COPD airway remodeling in rat model TGF-β1, VEGF content, adjust MMP-9/ TIMP-1 equilibrium state inhibition to fibroblasts and endothelial cells hyperplasia of collagen synthesis,is one of the mechanisms to delay or prevent airway remodeling in COPD disease.2. Siwutang and drugs on (Chuanxiong-angelica,Shudi-white peony) method can be nourishing and promoting blood circulation through, to improve the local micro environment,reduce inflammation,in order to improve the local mucosal immune function, is one of the mechanisms to delay or prevent airway remodeling in COPD disease.
