| Objective; To investigate the changes and significance on proportion of regulatory T lymphocytes in peripheral blood of rats model for chronic abacterial prostatitis based on successful establishment of the rats model.Method;①Twenty four similar shape male rats chosen, Wistar, about400g or so, were randomly and equally divided into four groups, such as high-dose-model group, low-dose-model group, vehicle group and blank control group.②17beta estradiol, after castration, was injected in abdominal subcutaneous of each rat for chronic abacterial prostatitis for30days. Rats in medium group were injected with sesame oil for30days too. Normal control group were fed as normal, and were given relevant treatment as other groups after30days.③After a month, blood and prostates of all rats narcotized by0.3%pentobarbital sodium were respectively collected and separated.④After their wet weight being weighed up with analytic balance, prostates were fixed with about4%formalin. Then the pathological change of prostate tissue, after being stained with Hematein Eosin, was observed with optical microscope.⑤Percentages of CD4CD25T lymphocytes, CD8CD28T lymphocytes and CD4+CD28+T lymphocytes from half fresh blood collected were separately determined by flow cytometry.⑥Expressions of IL-4and INF-y in corresponding-marked supernate sucked up from the other fresh blood after being immediately centrifuged were detected with enzyme-linked-immuno-sorbent assay. Data that were recorded was analyzed and disposed with SPSS17.0software.Result:(1)Wet weight of prostates from high-dose-model group and low-dose-model group, compared with that of sesame-oil control group and blank control group, dramatically deceased(P<0.01). Prostate tissues of model groups were observed obvious inflammation with the help of microscope, such as glandular epithelium atrophying, stroma having infiltration of inflammatory cells, and so on.(2)Compared with proportion of CD4+CD25+regulatory T lymphocytes in peripheral blood of rats from sesame-oil control group and blank control group, that of high-dose-model group obviously decreased(P<0.01) and that of low-dose-model group also declined(P<0.05). Besides, percentage of CD8+CD28-regulatory T lymphocytes in peripheral blood of rats from both high-dose-model group and low-dose-model group decreased compared to that of control groups(P<0.05). Change of proportion of CD8+CD28-regulatory T lymphocytes in experimental group positively correlated with the change of proportion of CD8+T lymphocytes. And proportions of CD4+CD25-T lymphocytes, CD4+CD28+T lymphocytes and CD8+CD28+T lymphocytes in peripheral blood of all rats from four groups were indiscriminate.(3)Expressions of IL-4and INF-y among four groups were nondistinctive.Conclusion;1) Rats model for chronic abacterial prostatitis was successfully established with17beta estradiol.2) CD4+CD25+regulatory T lymphocytes and CD8+CD28-regulatory T lymphocytes took part in the occurrence of chronic abacterial prostatitis, which would settle foundation for further study of chronic abacterial prostatitis. |
