Objective: To study the effect of astragalus polysaccharides (APS) with ileal transposition(IT) on Goto-kakizaki (GK) rats, then further to explore the influence of this kind of treatmenton fasting plasma insulin (FPI) and peptide Glucagon samples-1(Glucagon like peptide-1,GLP-1) respectively, meanwhile, analysis the relationship between astragaluaspolysaccharides (APS) and change of diet, drink, weight, blood glucose level of diabetic ratswhich are operated by ileal transposition (IT).Methods:20normal GK rats were randomly divided into two groups, with10rats in eachgroup. Rats in group A underwent the treatment that APS combined with IT, while rats ingroup B were just operated by IT. For all rats, their changes of diet, drink, and weight wouldbe recorded in this research. Recording the level of fasting plasma insulin and Glucagon likepeptide-1(GLP-1) in each group before and1,2,4,8,12weeks after operation.Result:1Within two weeks, compared with the preoperative status, the weight of all rats intwo groups decreased significantly (p<0.05); after two weeks, the weight of all rats increasedgradually, which were heavier than the preoperational level (p<0.05);12weeks afteroperation, the weight of rats in group A were much less than that of group B (p<0.05).2Thefasting glucose levels of all rats both in group A and group B were significantly reduced afteroperation (p<0.01); one week after operation, the blood glucose levels of the two groups ratsfell sharply, then achieved the lowest level; eight weeks after operation, the level of bloodglucose gradually recovered to the normal level but less than the preoperational level, thenkept this status until the end of observation period (p<0.01); there was no significantdifference in blood glucose level between two groups (p>0.05)3The level of fasting plasmainsulin of all rats both in group A and group B rose significantly after operation, whichcompared with preoperational level (p<0.05), as for the fasting insulin levels in rats, there wasno significant difference between the two groups of after operation (p>0.05);4The fastingplasma C-peptide level increased significantly after operation, there has the significantdifference in fasting plasma C-peptide level between preoperational period and after operation(p<0.05), there was no significant difference in fasting C-peptide level between two groupsrats after operation (p>0.05);5The fasting GLP-1level was significantly higher at each time point in two groups after operation than before operation (p<0.01), furthermore, the level ofgroup A was significantly higher than that of group B (p<0.01).Conclusion: IT can significantly reduce the blood glucose level in GK rats; the efficacy ofastragalus polysaccharides with ileal transposition treatment is better than IT operation; theeffective control of blood glucose level may be associated with the mechanism of increasingGLP-1secretion and insulin secretion.
