Objective1. The aim of this study was to determine if the sperm binding to the human zonapellucida (ZP) could be more selective for double stranded DNA than sperm afterswim-up.2. Study the influence of sperm DNA fragmentation index (DFI) and number ofsperm bound to ZP on the outcome of IVF-ETMethodsPatients as secondary infertility treated with the IVF-ET were selected, in whichthe female aged below35years old and had normal ovarian reserve. The DFI of theirsperm dope, sperm after swmi-up and sperm bound to ZP was detected by the spermchromatin dispersion(SCD) test, the DFI was compared among the threemethods.Study the influence of their DFI and number of sperm bound to ZP on theoutcome of IVF-ET.Results1. The DFI of the sperm dope, sperm after swmi-up and sperm bound to ZP were32.65%,16.84%,9.29%.There was statistically significant difference between eachtwo(P<0.05).2. The DFI of the sperm dope and fertilization rate had marked negative correlation, there was no significant correlation between the DFI of the sperm dope andhigh-quality embryo rate or implantation. The DFI of sperm after swmi-up andfertilization rate, high-quality embryo rate had marked negative correlation; there wasno significant correlation between the DFI of sperm after swmi-up and mplantation.The DFI of sperm bound to ZP and fertilization had no significant correlation; therewas significant correlation berween the DFI of sperm bound to ZP and high-qualityembryo rate or implantation. The number of sperm bound to ZP had significantcorrelation with fertilization high-quality embryo rate and implantation rate. Devidedthe patients into the clinically pregnant group and non-pregnant group, the DFI of thesperm dopec and sperm after swmi-up between them had no significant difference(30.2%VS36.8%,P=0.261;16.6%vs17.1%,P=0.843); there was statisticallysignificant difference in the DFI of sperm bound to ZP and the number of spermbound to ZP between them(6.9%vs10.1%,P=0.042;88vs67,P=0.046).Conclusion1. The DFI of sperm bound to ZP is significantly lower than the DFI of sperm afterswmi-up.2. The sperm DNA injury has bad influence on the outcome of the IVF-ET. Thenumber of sperm bound to ZP is closely related to its fertilizing capacity anddevelopmental potential after fertilization, so it can be used as an indicator to detectsperm function.
