| Objective:To investigate the effect of Zangyao Shibawei Ouqu Ointment on the eczemamodel level of serum Interleukin-4and Interferon-γ〠the pathology of skin lesions andinflammatory factor in rats.Preliminary study on the mechanism of Zangyao Shibawei OuquOintment for eczema.Method:According to weight, the74rats were randomly divided into2groups: with12rats in blank group and62rats in model group. One day before the sensitized,unhairing3cm×3cm on the back with depilatory paste, external application5%DNCB50μL on the backof rats sensitized the first time next day, unhairing3cm×3cm on the back too after2weeks,external application0.1%DNCB100μL on the back of rats excitation,one time one week andcontinuous4weeks, after sensitized3days, took2rats randomly from model group after themodel was achieved,cut their unhairing skin on back into pieces,10%formalin fixed,paraffin-embedded, section, HE staining, observed the pathological changes of eczemamodels with Light Microscopic, According to skin irritation test observation indexes, theeczema model of rats’ back was successful.The other60rats randomly divided into ZangyaoShibawei Ouqu Ointment low dosage group(0.14g/kg)ã€middle dosage group (0.21g/kg)andhigh dosage group(0.35/kg)ã€Binghuangfule groupã€model group(1g/kg),each group12rats,external application drugs once a day for10days continuously and observe everyday.Drawing materials from the4rats back of each group at3dã€7dã€10d,10%formalin fixed,paraffin-embedded, section, HE staining, observed the pathological changes with LightMicroscopic, and inflammatory cells counts with IPP image analysis software. At the sametime took few femoral artery blood of each group, isolated serum and preserved below20℃to determine the amount of cytokines(IL-4ã€IFN-γ)by ELISA.Results:1.Zangyao Shibawei Ouqu Ointment could improve epidermal keratinizationã€spinous cell layer thickeningã€apophysis prolonged pathological manifestations caused byDNCB in different degree; Each erythema were reduce or disperence,the score is decreasedthan model group, differences were statistical significance(P<0.05); Compared with modelgroup,lowã€middle and high dosage group(P<0.05),and compared with positive controlgroup,highã€low dosage group,they had statistical significance (P<0.05).Compared positive control group to middle dosage group,there was no significant difference (P>0.05),theefficacy were similar;2.Each treatment group dermis inflammatory cells were decreased thanmodel group, the differences were statistical(P<0.05);3. In the influence on the content ofIL-4and IFN-γ in the serum, compared with model group, IL-4andIFN-γ in the serum havedifferences were statistical other groups(P<0.05);There is no significant difference betweenthe middle dosage group and Binghuangfule group concerning the serum level of IL-4ã€IFN-γ(P>0.05). There was a significant difference between the low and high dosage groupand the control group(P<0.05).Conclusion:Zangyao Shibawei Ouqu Ointment could improve the erythema and dropsyof rats eczema model,which suggested that Ouqu Ointment have therapeutic effects foreczema,and with the improvement showed in a dose-effect relationship; After the eczemamodel was treatment with Zangyao Shibawei Ouqu Ointment,compared with blank group thelevel of IL-4is decreased and IFN-γ is incresed, Zangyao Shibawei Ouqu Ointment isobviously effective on the treatment of eczema,It could regulation the imbalance of T cellsubsets, the therapeutic mechanism of rats eczema model maybe related to regulation of Tã€Blymphocyte function,it suggested that Ouqu Ointment have the function of immune regulation.The therapeutic mechanism maybe related to Ouqu Ointment regulate induced inflammatoryã€anti-inflammation factorã€inhibit inflammatory cells. |
