This paper investigated the antioxidant action and the related antiaging mechanism with Drosophila melanogaster as animal model.At first, HPLC had been used in Phloridzin identification and purity determination. According to the standard curve and results of sample peak area, the purity of phloridzin is73%.Afterwards, antioxidant activity of Phloridzin in vivo had been researched. Firstly, established a model of normal flies and fed the flies with different dosages of phloridzin. Flies were transferred to vials containing fresh medium every3days and deaths were scored. At25day or40day, SOD and CAT activity and MDA content were measured.Accordingly, Total RNA of the fruit flies at25day was extracted and isolated, Real-time PCR amplification assay was carried out to quantify the mRNA level.The results showed that The present study demonstrated that phloridzin showed a lifespan extension effect of D. melanogaster in a dose dependent manner.SOD and CAT activities were improved significanty(p<0.01),meanwhile the content of MDA reduced.Results of Real-time PCR amplification assay showed that phloridzin supplement could upregulated Gene expression of CuZnSOD, MnSOD and CAT.Secondly, establish a model of flies exposed to ultraviolet (UV) rays. Longevity test and biochemical analysis methods was used to discuss the protected effect and the mechanism of phloridzin about the damage of oxidative damage induced by UVR. Phloridzin treated flies caused a significant decrease in MDA content and increase in SOD and CAT activities in response to the lifespan extension.Their supplementation can rescue the short life span induced by ultraviolet (UV) rays.Thirdly, establish a model of hyperlipidmia flies fed the flies with different dosages of phloridzin.10%lard was added to the normal medium as the high fat dietry to discuss the protected effect of phloridzin on the fat-induced oxidative damage in Drosophila melanogaster. Fat accelerated the ageing process of D. melanogaster, while phloridizn reversed partially the fat-induced mortality.Finally, hydrogen peroxide treatment and paraquat treatment was used to exmine the resistance of flies agaist oxidantive stress.In acute experiment, the supplementation of phloridzin prolonged the survival time of fruit flies in in a"dose-response relationship.The present study demonstrated that phloridzin possess strong antioxidant activity in vivo.
