Objective:To explore Impacts and action mechanism of Mieyoutang on TLR2and NF-κBP65in H. Pylori-Associated Gastritis mice gastric mucosa tissues.Methods:(1)BALB/c mice were randomly assigned to normal control group, model group, gastric triad group. miey-outang group.(2) developed a model of HAG BALB/c mice Spleen-stomach Damp-heat Syndrome by composite factors (high sugar and fat food+hot and humid environment+H.Pylori). The mice were observed drinking, diet, stool, weight, rectal temperature conditions and general behavior change.(3) After three days Successful modeling, Model group and blank control group were distilled water14days,the other groups were distilled Corresponding drugs.(4) To test Serum HP antibody by Colloidal gold method,To observe the pathological morphology of gastric mucosa under microscopy. The immunohistochemical assay was adopted to detect the protein expressions of nuclear transcription factor TLR2and NF-κBP65. Quantitative PCR was adopted to detect the gene content of TLR2mRNA and NF-κBP65mRNA.Results:(1)Colloidal gold method tested positive, HE staining under light microscope observated mucosal inflammation, model group and the control group there was a significant difference,(p<0.01).(2) Compared with normal control group, the expressions of TLR2and NF-κBP65and the gene content in gastric mucosa of mice increased in model group(p<0.05),compared with model control group, the expressions ofTLR2and NF-κBP65and the gene content in gastric mucosa of mice decreased in mieyoutang group(p<0.05), compared with normal control group,the expressions of TLR2and NF-κBP65and the gene content in gastric mucosa of mice increased in mieyoutang group(p.>0.05).Conclusion:Mieyoutang can regulate the expressions of TLR2and NF-κBP65in gastric mucosa of mice,alleviates gastric mucosal inflammation. Mieyoutang treats HAG probably through down-regulate the expressions of TLR2, inhibit the activation of NF-κBP65, thus hinder the activation of NF-κB, furthermore perform treating effect.
