| Objective:The study by makig ulcerative colitis model rats observe the influence of the Classical Prescriptions Gancao Xiexin Decoction on the pharmacodynamics of rats and NF-κB signaling pathway and anti-inflammation cytokine(IL-10).To discuss the underlying mechanism of action of the treatment of ulcerative colitis,the study provide ideas and theoretical basis for clinical application Classical Prescriptions in treatment and prevention ulcerative colitis.Methods:The ulcerative colitis model reference Jiangxueliang’s law and a little improvement was established by composite method with2,4-Dinitrochlorobenzene (DNCB) and acetic acid.90Wistar rats were feeding adaptability a week and divided randomly into9groups:10rats per group,They are Xiexin Decoction high dose gavage group, Xiexin Decoction medium dose gavage group, Xiexin Decoction low dose gavage group, Xiexin Decoction High dose clyster group, Xiexin Decoction Medium dose clyster group, Xiexin Decoction low dose clyster group, western medicine group, model group, normal group.Normal group free diet.Model group rats were given by gavage once a day with distilled water4ml.Western medicine group rats were given by gavage once a day with sulfalazine(SASP)0.45g/kg. Xiexin Decoction low dose group were respectively given by gavage and clyster once a day with Gancao Xiexin decoction lml/100g. Xiexin Decoction medium dose group were respectively given by gavage and clyster twice a day with Gancao Xiexin decoction lml/100g. Xiexin Decoction high dose group were respectively given by gavage and clyster twice a day with Gancao Xiexin decoction1.5ml/100g.The medication was given14days. At the end of the experiment, the disease activity index(DAI) was evaluated. The colon mucosa damage index(CMDI) and tissue damage index(TDI) were evaluated.Interleuikin-10(IL-10) in serum concentration was detected in the way of enzyme-linked immunosorbant assay(ELISA).And the positive expression of nuclear factor kappa Bp65(NF-κBp65) in the colonic mucosal tissue was detected by means of immunohistochemical.Result:From the phamacodynamics, model group compared with normal control group, it had significant difference (p<0.01).No significant differences were found in the treatment groups (p>0.05). The serum concentration of IL-10was decreased significantly in the model group, and compared with the normal group, it had significant difference(p<0.01). The serum levels of IL-10was increased significantly in the treatment groups, and compared with the model group, it had significant difference (P<0.01). Western medicine group compared with Xiexin Decoction high dose group, it had statistical significance (p<0.05). The nuclear factor kappa Bp (NF-κB) was increased significantly in the model group, and compared with the normal group, it had significant difference (P<0.01). Most of the NF-κB positive staining is given priority to karyon. The positive expression of NF-KBp65was decreased significantly in the treatment groups, and compared with the model group, it had significant difference (P<0.01).Western medicine group compared with Xiexin Decoction high dose group, it had statistical significance (p<0.05).Conclusion:From the phamacodynamics, Gancao Xiexin decoction and the SASP were effective in relieving the symptom of ulcerative colitis model rats, reducing colon mucosa damage and promoting ulcer healing to different extents. But it coundn’t prove the efficacy of Gancao Xiexin decoction was better than the SASP. The serum concentration of IL-10and the positive expression of NF-κBp65showed the dosage of Gancao Xiexin decoction and the efficacy had some relationship. The more the dosage, the better the efficacy. |
