Objective:(1)Through the bilateral common carotid artery ligation rats, establishinganimal model of vascular dementia,and observe the behavior of chronic cerebralischemia in rats after change.(2)Research on different time points after chroniccerebral ischemia rats Nogo-A change of expression, and discusses Nogo-A in theoccurrence of vascular dementia rats and the significance in the process ofdevelopment.Methods:(1) Choose40wistar rats were randomly divided into model group and thecontrol group. Application of model group rats permanent bilateralcommoncarotidarteryligation method.(2vo) prepared by chronic forebrain ischemiaanimal model of vascular cognitive impairment in the control group only procedureand no bilateral common carotid artery ligation.(2) Using Morris water maze test ofeach group rats before and after operation, behavioral changes and score.Comparison before and after surgery and the differences between the groups.(3)After15days,30and60days respectively take10rats in each group, beheaded inthe brain. Line will be made part of the brain tissue slices, Elisa dyeing, statisticNogo-A change of expression.Results:Mdel group rats after15days,30and60days learning and memory abilitysignificantly decreased compared with control group,15days is most obvious.(2)Elisa staining showed model group rats Nogo-A more apparent, with increased15days before30days and60days rise more slowly.Conclusion:(1) Chronic cerebral ischemia can make the learning and memory of ratsbehavior ability to drop, chronic cerebral ischemia caused by ligation of bilateralcommon carotid arteries could be successfully establish a vascular dementia modelin rats.(2) Chronic cerebral ischemia caused by increasing Nogo-A expression maybe the nerve regeneration after injury of adult mammalian barrier one of the important reasons.
