Plant pigments as one kind of natural edible pigment have a long history. They are not only nuisanceless, but also have the function of health care. Because the artificial chemical pigments have simple process and low price, so they are popular with people for a long time. However, the artificial chemical pigments are bad for people’s health. Hence, it is great urgency to study natural pigment in terms of reducing cost and simplifying the technology. In this paper, we study the curcumin through the aspects as are mentioned above.Firstly, we study the test method of curcumin. We mainly use ultraviolet spectrophotometer and HPLC to test the curcumin and draw the standard curve of ultraviolet spectrophotometer and HPLC, which is important for the subsequent test. We also study the liquid semipreparative way of curcumin that is the foundation of preparation of high purity of the samples.Then, we study the influence of aqueous two-phase on separation and purification of curcumin. Through changing the PH, temperature and percent of (NH4)2SO4 constantly, we get the best condition of split-phase. The conclusion as follows:the temperature and PH have little influence on aqueous two-phase of (NH4)2SO4, which is convenient to control experiment. The percent of (NH4)2SO4 affect the split-phase of aqueous two-phase directly, so we must control the percent of (NH4)2SO4 strictly.Besides, macroporous resin affect the purge process through wet packing method. We let the concentration of sample is 0.7mg/ml and observe the result of experiment on the conditions that the sample weight are 50ml,35ml,20ml. Consequently, we could find that we can get the best result when the sample weight is 35ml and the purity of curcumin is 22%.Last but not least, we compare the process of extraction under some conditions such as conventional stirring extraction, single frequency ultrasonic extraction and dual-frequency ultrasound extraction. Above all, we explore the best result of single factor experiment. After that, we do orthogonal experiment and get the best result of experiment. Finally, we verify the result through orthogonal experimental validation. We find that the best method is dual-frequency ultrasound extraction at last. We get the final purity of curcumin is22% through combining the aqueous two-phase and purification of macroporous resin.
