With the world becoming more and more aging population, age-related disease prevention and treatment of medical research has become a hot spot.Vaginitis is a very common gynecological disease and troubled by the majority of female friends.vaginitis is also a serious hazard common disease in the elderly women. Menopausal syndrome is one of the major clinical manifestations. Developing natural, healthy, broad-spectrum, highly efficient, non-toxic side effects of modern health gynecological diseases topical drugs is the main trend of drug research.The study is based on traditional Chinese medicine prescription, through a wide range of information access and market research, established the basis for the treatment of senile vaginitis prescription-angelica, sophora alopecuroides, by Chinese medicine theory, and used modern technology and formulation preparation for traditional Chinese medicine angelica bolt technology research and development.The main contents include the following five aspects:1).The research of extraction of angelica, paste rate and ferulic acid as indicators, use of percolation method, the investigation of factors are the amount of ethanol, ethanol concentration and soaking time for the three factors and three level orthogonal experiment, and the best extraction process is a 12-fold volume of liquid ratio,70% ethanol, soaked 4 hours percolation extraction process; The research of extraction of shikonin, the paste rate, total naphthoquinone content, and shikonin content index as indicators, percolation method with immersion time, ethanol concentration, solid to liquid ratio were investigated factors, and did the three factors and three levels orthogonal experiment, the best extraction process with 8 times the amount of liquid ratio,90% ethanol, soaked for 4 hours percolation;extraction process of sophora, respectively, the extract rate, total alkaloids, matrine in matrine and oxidation as indicators, using reflux method, as the reflux time, ethanol concentration, the ratio of liquid to material factors investigated, doing the three factors and three level orthogonal experiments, to determine the amount of liquid to solid ratio of 12 times,70% ethanol, reflux extraction twice, each time 1 hour extraction of bitter beans the best. 2). When angelica, sophora., lithospermum extract equivalent to raw herbs 3:1:1; 1:1:3; 1:3:1 three ratio, to research anti-inflammatory activity. Using xylene-induced mouse ear swelling test method, with metronidazole as a positive control, the results show that the swelling of three groups were 1.57,3.27,2.71, and blank control group was significant differences in type, in which the anti-inflammatory effect of high pain group was significantly better than the positive group. Therefore, the preparation process of follow-up to high pain group as prescribed ratio.3). To develop a HPLC method for determination of matrine, oxymatrine, ferulic acid, L-shikonin andβ,β-dimethylacrylshikonin in compound recipe preparation of Ziguizishuan. The method was proved to be credible and accurate.All the compounds showed good linearity (r>0.9996) in the range of the test concentrations, and the average recoveries of the method was between 96.92% and 102.22%, RSD<3.12%. it can be used to compound the quantitative quality control of Ziguizishuan5). The TLC research of angelica compound suppository formulations. Three batches of angelica, Lithospermum erythrorhizon, Sophora alopecuroides L and the corresponding bean ferulic acid, shikonin andβ,β-dimethyl acrylic acid aka ning, matrine were as the reference analysis, the results showed that all three batches of preparations containing the reference elements, and established method is simple, reliable, and reproducible, and can be used for qualitative preparation of Ziguizishuan quality control.
