Objective:Nickel carbonyl had produced hundreds of acute toxicity accident since it had been found until now, and it is one of the chemical metallurgical industrial production of highly toxic compounds. Carried out the research of cytology for the drug efficacy of acute nickel carbonyl poisoning in rat would provide theoretical guidance for clinical first aid and development of new clinical drugs.Methods:SD rats groups include of control group, carbonyl nickel group, methylprednisolone (20mg/kg), DDC (100mg/kg), sodium selenite (10μmol/kg), shenfuhuiyang injection (0.25ml/kg) and and methylprednisolone with DDC (20mg/kg+100mg/kg). All rats inhaled passively 250 mg/m3 carbonyl nickel for 30 minutes except for control group. They were injected in abdomen at 4 or 30 hours after poisoned, and then were observed at 3 or 7 days, each group has 10 rats. Application methods of Single Cell Gel Electrophoresis (SCGE) and Laser Scanning Confocal Microscope were used to detect the repairment of cell DNA and free calcium in cell of acute nickel carbonyl poisoning in rat lung, liver, spleen, lymphocytes.Results:1. Analysing the repairment of comet tail and olive tail of acute nickel carbonyl poisoning in rat lung, liver, spleen, lymphocytes by SCGE and software of CASP, which showed as followed. Compared to the poisoned control group, methylprednisolone, DDC, sodium selenite, shenfuhuiyang injection and methylprednisolone with DDC shown significantly therapeutic effects for rat cells exposed acute carbonyl nickel poisoning (P<0.05). Compared to the normal control group, the damage repair of DNA, mitochondria and other organelles by methylprednisolone and DDC and methylprednisolone with DDC would approach to pre-poisoned levels (P>0.05), with the treatment efficacy was much better at 4 hours injection after poisoned than that after 30 hours (P<0.05). However, the treatment efficacy was much better at 3 days observation after poisoned than that at 7 days in DDC (P<0.05).2. Analyze the repairment of free calcium in cell of acute nickel carbonyl poisoning in rat liver, spleen, lymphocytes by Laser Scanning Confocal Microscope, it showed the effect were better in methylprednisolone and DDC than in other groups (P<0.05), and the treatment efficacy was much better at 4 hours injection after poisoned than that after 30 hours (P<0.05).Conclusions:1. There were some treat effective by the lung, liver, spleen and lymphocytes cells DNA damage after exposed to the acute carbonyl nickel in methylprednisolone, DDC, sodium selenite and shenfuhuiyang injection and methylprednisolone combined with DDC, and the effect were better in methylprednisolone and DDC than in other groups, and the treatment efficacy was much better at 4 hours injection after poisoned than that after 30 hours.2. Methylprednisolone and DDC if that used early could promote the repairment of free calcium in cell of acute nickel carbonyl poisoning in rat liver, spleen, lymphocytes.
