Salvia chinensis Benth is the all parts of Sage herb. This genus has anti-tumor, anti-viral,anti-inflammatory and other significant pharmacological activity. As an effective anti-tumorherb, Salvia chinensis Benth have been used widely in folk, but its active ingredients arerarely reported. The studies on the chemical constituents and pharmacological activities, canprovide certain theory basis for making full use of the plant resources and researching anddeveloping in depth.Firstly, the extraction of Salvia chinensis Benth piece was extracted three times with70%ethanolasmicrowave-assisted. After suspension procedure by adding water, different polarsolvents were used for the five fractions of petroleum ether, chloroform, ethyl acetate andn-butyl alcohol and aqueous phase were gotten. Meanwhile, the effect of anti-colon cancerwas carried out for the five fraction extracts of Salvia chinensis Benth. The obtained resultswere that the chloroform and ethyl acetate extraction have significantly stronger anti-canceractivity than the other phase extraction.The chloroform fractions of Salvia chinensis Benth ethanol extract was subjected toseparate and purify its anti-colon cancer active constituents by large silica gelcolumn,polyamide column, ODS open column and sephadexpreparation column, combinedwith the screening experiment for active constituents. Threecompounds were gotten from thecholoroform fractions. Using mass spectroscopy,1H and13C nuclear magnetic resonancespectroscopy and comparison with literatures, theyare determined to be ursolic,sitosterol andoleanolic acid.The ethyl acetate extract of Salvia chinensis Benth ethanol extract was subjected toseparate and purify its anti-colon cancer active constituents by polyamide column, silica gelcolumn, ODS open column and sephadex preparation column, combined with the screeningexperiment for active constituents. Two compounds were gotten from the ethyl acetate extract.Using mass spectroscopy,1H and13C nuclear magnetic resonance spectroscopy andcomparison with literatures, the two monomeric compounds were identified to be: vanillinand resveratrol.Guidedby antitumor activityand investigationon their eftcetsinvitro,ursolic,sitosterol,oleanolic acid,vanillin and resveratrolwere isolated from Salvia chineses Benth. Theanti-colon cancer activity of the five compounds were evaluated through cell activityexperiment in vitro, when the five compounds contact with the Caco-2cells of human bodyfor72hours, the EC50were30.49μg·mL-1、29.17μg·mL-1、35.58μg·mL-1、39.25μg·mL-1and 22.63μg·mL-1respectively.When the five compounds contact with the HepG2cells of humanbody for72hours, the antitumor effective were not as good as the anti-colon ettective.
