| Objectivethe Salvia chinensis Benth was extracted to different compounds for acquire the active situs with anti-HIV effect in vitro.Methodsthe extraction of ethanol,from Salvia chinensis Benth carried out 75%industrial alcohol,which was extracted by using Petroleum ether,chloroform,ethyl acetateand butanol.To acquire the crude extractive of chemical composition against HIV from the chloroform of Salvia chinensis Benth.The crude extractive of effective compounds were isolated from chloroform of the Salvia chinensis Benth by silica gel column chromatography,Thin layer chromatography,vanillin coloration,and high performance liquid chromatography,nuclear magnetic resonance(NMR).CEMxl74 with HIV was used to detected the effect of herb,abserved CPE under microscope and detectd HIV RNA relative amount and the supernatant of the viral load of HIVRNA by RT-PCR as three indicators,which is used to detect the extraction whether has anti-HIV effect in vitro.the statistical method is that using ABI7300 quantitative PCR instrument carried software analysis to deal with the file from the operation of quantitative PCR,and then using Excel build database,SPSS17.0 statistical software for statistical analysis and test methods using one-way analysis of variance,if the variance is standard then to use the LSD method analysis,if the variance is not and then used by Kruskal-Wallis Hmethod analysis,besides,the test standard α=0.05,GraphPad Prism 5.0 for Graphic Software.ResultsFor crude extractive from Salvia chinensis,pharmacological experiment of HIV infect the cell in vitro,which showed that concentration of chloroform was 20μg/ml is effective by observe the result of CPE,meanwhile,compared with the model group,the results of intracellular HIV RNA relative amount of RT-PCR showed that concentration of 20μg/ml from chloroform of the Salvia chinensis Benth in the treatment group is effective.(P<0.05);and compared with the positive control group(P=0.128>0.05);There is no difference.compared with the model group,the results statistic of the supernatant of RT-PCR showed that oncentration of 20μg/ml from chloroform in the treatment group is effective(P<0.05),and compared with the positive control group(P=0.611>0.05),There also is no difference.The results showed that the effective crude extraction sites were eluted by silica gel column chromatography,thin layer chromatography,vanillin coloration,and high performance liquid chromatography and nuclear magnetic resonance identification structure,which was the anti-HIV active compounds ursolic Acid,Besides,for chloroform of the Salvia chinensis Benth,pharmacological experiment of HIV infect the cell in vitro,which showed that 22-23 and 5-6 was effective by observe the result of CPE,meanwhile,compared with the model group,about intracellular HIV RNA relative amount and istatistic of the supernatant,the results of RT-PCR showed that the 22-23 and 5-6 from chloroform of the Salvia chinensis Benth in the treatment group is effective.(P<0.05);and compared with the positive control group(P>0.05),There is no difference.Conclusionthe chloroform of the Salvia chinensis Benth has an inhibitory effect on HIV-1.The 22-23,ursolic Acid,and 5-6fraction is the main active ingredient from chloroform of the Salvia chinensis Benth inhibit HIV in vitro. |
