Objective:Through by observing the Salvia chinensis Benth.Polysaccharides(SBP) the effection on the inhibition and apoptosis of sw480 colon cancer cells, thus, we explored the mechanism of SBP inducing apoptosis of colon cancer SW480.Methods:(1)Get the SBP through aqueous extraction method.(2)Detected the SW480 cells proliferation at different times with different concentrations of SBP by MTT, screening out a suitable concentration gradient and time, then with the best conditions the induction of apoptosis of SW480 cells was determined by Annexin-FITC/PI assay.(3)Tested the expression of PI3 K,P-AKT,AKT,Caspase-3 in SW480 cells by Western Blot.Result:(1)The SBP inhibit the proliferation of SW480 colon cancer in a dose-time dependent effection relationship. In SW480 colon cancer 24 h,48h, 72 h, 0.0mg/ml~3.0mg/ml the SBP, the cell growth inhibition rate from 5.1±4.3% to 69.4±2.6%, 4.6±5.2% to 79.4±3.4%, 4.7±4.8% to86.5±2.3%(P<0.05), respectively,.(2)Annexin-FITC/PI assay showed that the SBP promoted the apoptosis of SW480 colon cancer. In SW480 cells 72h, 0.0mg/ml~2.0mg/ml the SBP, the rate of apoptosis from10.83±0.61% to 45.80±6.01%, P<0.05.( 3) In SW480 colon cancer72 h,Western Blot showned that expression of PI3 K,P-AKT in SW480 cells were down-regulated after addition of the SBP, the expression of caspase-3 was a opposite trend, while made no change in AKT.Conclusions: The SBP could inhibit the proliferation and promote the apoptosis in SW480 cells, and the molecular mechanism may reduce the expression of PI3 K and control AKT phosphorylation through PI3K/AKT signaling pathway.
