Effect Of Yiqi Jiedu Method On Tumor Bearing Nude Mice Tumor Inhibition And Inhibition Experimental Study On Gastric Cancer Cell Proliferation And Apoptosis

Objective： To study the effect of Yiqi Jiedu method on tumor bearing nude micetumor inhibition and inhibition of gastric cancer cell proliferation and induce apoptosis,to explore the mechanism of this method, in order to provide theoretical foundationand basis for the clinical application of Yiqi Jiedu Decoction in treatment of gastriccancer.Methods：the effect on gastric cancer cells in vitro and in vivo experiments to observethe effect of Yiqi Jiedu method. In vivo experiment of Yiqijiedu formula based on thetheory of water extraction and alcohol precipitation process preparation, madestabilizer type; human gastric cancer cell lines SGC-7901, MGC-803were cultured invitro, using MTT assay on human gastric cancer cell line by the drug susceptibility test,according to the48h pre experiment half inhibitory concentration IC50determinemedicine,, low dose, observation of cell morphology and cell activity under theinverted phase contrast microscope, cells were measured by ELISA absorbance oddetection instrument, the cell inhibition rate; by immunofluorescence technique wasused to observe the expression of CyclinD1, Bcl-2, Caspase-3inside cells, threeproteins; to observe the effect of Yiqijiedu method on gastric cancer cell proliferation,apoptosis from the cell level. In vitro by constructing gastric bare orthotopictransplantation tumor model in mice, to observe the effect of Yiqi Jiedu Decoction onbody weight in mice in bare the orthotopic transplantation tumor of tumor weight andtumor bearing mice, and immunohistochemistry, Westernblot technology, from themolecular level to investigate the Tiger Decoction on gastric effects of nude miceorthotopic transplantation tumor cell proliferation, apoptosis associated protein CyclinD1, Bcl-2, Caspase-3expression.Results：in vitro,(1) in the MTT experiment, the experimental group and the controlgroup had significant difference, P <0.05; traditional Chinese medicine on humangastric cancer cell line SGC-7901, MGC-803showed cytotoxicity; traditional Chinese medicine dose and cell apoptosis in gastric cancer cells is related to traditional Chinesemedicine, namely the higher dose on gastric cancer cell proliferation inhibition effectis more strong, cell viability was worse;(2) immunofluorescence test, CyclinD1, Bcl-2,Caspase-3on human gastric carcinoma cell line SGC-7901, MGC-803significantly inthe expression of Yiqi Jiedu Decoction group and control group, P <0.05; Caspase-3inYiqi solution under the influence of drug toxicity increased positive rate, were higherthan the control group; but CyclinD1, the Bcl-2rate decreased in the effect of YiqiJiedu Decoction drug intervention positive expression, and were lower than the controlgroup. In vivo experiment,(1) the tumor bearing nude mice weight and tumor weightof the statistics, found that the traditional Chinese medicine group tumor weight lowerthan NS group, and the difference was statistically significant, P <0.05;(2) in theimmunohistochemical and Westernblot experiment, CyclinD1, Bcl-2expression inChinese medicine group was significantly lower than the control group, the expressionof Caspase-3in traditional Chinese medicine group was significantly higher than thatin control group, and the difference is statistically significant, P <0.05.Conclusion：The decoction and gastric cancer is closely related to cell apoptosis, themedicine has the inhibitory effect on gastric cancer cell proliferation, inhibition isproportional to the concentration of drug. Herbs on gastric cancer cell lines fromdifferent sources, different sensitivity of different drugs, tumor inhibition andinhibition of gastric cancer cell proliferation activity was related to. The progress andmetastasis of gastric carcinoma and expression of CyclinD1, Bcl-2, Caspase-3areclosely related to the expression of CyclinD1, Bcl-2and gastric cancer cellproliferation is related to cell apoptosis, Caspase-3and gastric cancer is related to.Based on gastric cancer cells and in tumor bearing nude mice performed better thanMTT experimental study of color, immunofluorescence, immunohistochemistry andWesternblot Qi Disintoxication, prompting Yiqijiedu method can inhibit proliferation,promote apoptosis of gastric cancer cells, has inhibitory effect on tumor bearing nudemice.
