Objective:The purpose of this project is designed to observe the change of prostate index,basic fibroblast growth factor (bFGF) and transforming growth factor-β1(TGF-β1) of variousexperimental groups to evaluate the effect of Bushen Huoxue Tonglin Decoction, theformula for reinforcing kidney,activating blood and treating stranguria,on bFGF andTGF-β1of model rats with benign prostatic hyperplasia (BPH) and to probe the possiblemechanism of Bushen Huoxue Tonglin Decoction for benign prostatic hyperplasia(BPH),andto provide scientific theory and experimental evidence for the treatment of benign prostatichyperplasia (BPH) in Traditional Chinese Medicine.Methods:72male Wistar rats with3months old, weighed and numbered, were randomlydivided into6groups,12rats in each group, the sham-operation group, the model group, thefinasteride group (positive control group), the low dose Bushen Huoxue Tonglin Decoctiongroup, the middle dose Bushen Huoxue Tonglin Decoction group, the high dose BushenHuoxue Tonglin Decoction group. Set up the model after rats received food and water adlibitum in1week of acclimation. Each group were anesthetized by10%chloralhydrate(3.5mg/kg) through intraperitoneal injection, operationed under aseptic conditions,excised bilateral testicular through a transperitoneal approach,the sham-operation grouponly dissociate bilateral testicular after anesthesia.Recovery7days after operation, thesham-operation group was given10ml/kg normal saline(NS) instead of inject testosteronepropionate.The rats in the rest5groups were treated with subcutaneous injection oftestosterone propionate5mg/kg per day to make model.Once modeling was begin, the modelgroup was given10ml/kg normal saline(NS),the finasteride group was given0.8mg/kg(equivalent to10times of clinical dose,the solution was made up by5mgfinasteride dissolved in10ml NS).5ml/kg of Chinese drugs solution(1.5g/ml) was given torats of the low dose Bushen Huoxue Tonglin Decoction group(equivalent to5times ofclinical dose),10ml/kg of Chinese drugs solution(1.5g/ml) was given to rats of the middle dose Bushen Huoxue Tonglin Decoction group(equivalent to10times of clinical dose),15ml/kg of Chinese drugs solution(1.5g/ml) was given to rats of the high dose BushenHuoxue Tonglin Decoction group(equivalent to15times of clinical dose).All drugs wereadministered by intragastric administration(i.g.) once a day,30days in total, the rats wereweighed weekly to adjust the doesage.24hours after the last administration,the rats weresacrificed and taken out the prostate tissue,then packet marking. Weighed the rat prostates byelectric-light analytical balance to get the wet mass of the rat prostates. At the same time,making specimens. Each group was analyzed and observed the changes of specimens incellular morphology under light microscope, detected the bFGF、TGF-β1expressions ofprostate tissues by method of immunohistochemistry.Result:The prostatic index and bFGF expression levels in3Chinese medicine treatmentgroup were significantly lower than those in the model group, the expression level of TGF-β1were significantly higher than those in the model group, especially for the high dose BushenHuoxue Tonglin Decoction group.The comparisons among the high dose Bushen HuoxueTonglin Decoction group, the middle dose Bushen Huoxue Tonglin Decoction group, thelow dose Bushen Huoxue Tonglin Decoction group and the finasteride group showedstatistical significant difference (P<0.01).Prostate tissue of rats in the model group showed obvious pathologic changes, thepathologic changes of the middle dose Bushen Huoxue Tonglin Decoction group, the lowdose Bushen Huoxue Tonglin Decoction group and the finasteride group was slighter thanthat of the model group, the high dose Bushen Huoxue Tonglin Decoction group wassignificantly lower than that of model group.
