| Methods:(1) Analysis Xiayuxue soup liquid alcohol extract active ingredients.(2) Cultured human gastric cancer SGC-7901cells and established xenograft models.(3) The nude mice were randomly divided into control group, the group Xiayuxue soup, fluorouracil (5-Fu group) and Xiayuxue Tonga fluorouracil combined treatment group (hereinafter referred to as joint group)10, group administration.24hours after the last administration, remove the orbital blood nude mice experiments, mice were killed, peel intact tumor mass and weighed to calculate the inhibition rate.(4) Detection the blood of the nude mice, liver and kidney function and tumor necrosis factor (TNF-a).(5) HE staining were observed in tumor tissue morphology.(6) Impact on Bcl-2, Bax and Caspase-3protein expression in tumor tissue immunohistochemistry assay Xiayuxue soup.Results:(1) Xiayuxue soup alcohol extract containing laetrile, allantoin, aloe emodin and chrysophanol, but the content of Aloe smaller, mainly for the first three.(2) Xiayuxue soup group on SGC-7901cells in nude mice xenograft tumor inhibition rate of36.5%,5-Fu group inhibition rate was44.0%, combined administration group inhibition rate was50.5%. There were significant differences (p<0.05) three groups.(3) Blood and liver and kidney function results are shown:5-Fu group of white blood cells and platelets appear nude universal values decreased kidney function related indicators increased, compared with the control group, the difference was statistically significant (P<0.01); hemoglobin values Xiayuxue soup group increased liver and kidney function related indicators lower, the difference was statistically significant (P<0.01) compared with the control group; Xiayuxue soup group and combined group, the white blood cells in nude mice, platelet levels relative to5-Fu group was significantly increased, and decreased kidney function-related indicators, the difference was statistically significant (P<0.05).(4) TNF-a are shown:Xiayuxue soup group, and TNF-a levels in the combination group was significantly higher compared with the control group, the difference was statistically significant (P<0.05).(5) The results of immunohistochemistry shows:Bcl-2protein expression Xiayuxue soup group was significantly lower, while Bax protein expression was significantly increased, there was significant difference compared with the control group; whereas no significant effect on the expression of Caspase-3no statistically significant difference compared with the control group. Its down Bcl-2, Bax up-regulated the expression of the role of5-Fu group with no significant difference in comparison, but significantly lower than the combined group.Conclusions:(1) Xiayuxue soup can SGC-7901significantly inhibited tumor growth in nude mice. Improve anemia, reduce the5-Fu-induced liver and kidney dysfunction and toxicity of myelosuppression (2) Xiayuxue from soup to some extent.(3) Xiayuxue soup can raise body levels of TNF-a, causing tumor necrosis and hemorrhage enhance immune function.(4) Anti Xiayuxue soup SGC-7901xenograft tumor effect may increase Caspase-3protein, Bcl-2protein and decrease the ratio of Bcl-2/Bax related, and co-administered with5-Fu, the role is more obvious. |
