Objective:Observed pathological and ultrastructural changes of lung tissue when traumatic hemorrhagic shock secondary lung injury in rats in dry heat desert environment, simultaneously detect the changes of apoptosis index, the total amount of protein in the lung lavage, wet-dry weight ratio, oxidative stress-related indicators and concentration of important inflammatory factor in serum, cyclooxygenase-2(COX-2) and inducible nitric oxide synthase (iNOS) mRNA of tissue and other,to investigate the mechanisms between indexes and secondary lung injury occurring. Methods:140male SD rats were randomly divided into the normal temperature environment traumatic hemorrhagic shock group (normal temperature group) and the dry heat traumatic hemorrhagic shock groups (dry group), respectively, each groups were randomly divided into7subgroup:the control group, after shock0h,0.5h,1h,1.5h,2h,3h group,10rats in each subgroup, establish the model, collect specimen, observe the pathological and ultrastructure changes of lung tissue, apoptotic index of lung tissue by TdT-mediated dUTP nick-end labeling (TUNE), detect the total amount of protein in the lung lavage and oxidative stress-related indicators, Enzyme-linked immunosorbent assay (ELISA) was used to detect serum concentrations of serum important proinflammatory cytokine.detected expression levels of lung tissue iNOS, COX-2mRNA by Real-time PCR. Results:Pathology change of dry group is more serious than the normal temperature group, alveolar epithelial cells edema, mitochondrial cristae, disappeared, the vascular basement membrane are destroyed,the invasion of inflammatory cell, apoptotic index, wet-dry weight ratio, the total amount of protein in the lung lavage are higher than normal temperature group, peak value as early; oxidative stress response more serious than the normothermia group, concentration of important inflammatory factor in serum and expression levels of lung tissue iNOS, COX-2mRNA are higher than that in normal temperature group, the peak appeared early; correlation analysis showed that between all index and pathological score of lung injury there is a correlation,and also exist between some indexes.Conclusion: When traumatic hemorrhagic shock secondary to lung injury in the dry heat desert environment, the lung injuries was serious and earlier than under the normal temperature ambient, cell apoptosis, oxidative stress-related indicators, important inflammatory factor in serum, expression levels of lung tissue iNOS, COX-2mRNA and other play a very important role in dry and hot desert environments by the process of traumatic hemorrhagic shock secondary to lung injury.
