Avian influenza virus poses a serious threat to the poultry and human. Due to the virus variability and drug resistance, there is no effective prevention and treatment measures. Therefore, researching and developing new anti-HPAIV drugs are imperative. Screening safe and effective anti-HPAIV compounds from chinese herbal medicines provides a large opportunity for the development of anti-HPATV drugs. In our previous studies, we found that the extracts of Cudrania cochin-chinensis had significant activity against the highly lethal human H5N1influenza virus.In our ongoing study, the active fractions were subjected to a series of separation and purification steps to abtain thirty six compounds. Thirty three of them were elucidated by spectroscopic methods and physicochemical properties as1,3,6-trihydroxy-7-methoxyxanthone (GJ-1),1,3,6,7-tetrahydroxy-8-prenylxantho-ne (GJ-2),1,3,5-trihydroxy-4-(3’-hydroxy-3’-methylbutyl) xanthone (GJ-3),1,3,6-trihydroxy-4-prenylxanthone (GJ-4),1,3,6,7-tetrahydroxyxanthone (GJ-5),1,3,5,8-tetrahydroxyxanthone (GJ-6),1,3,6-trihydroxy-4-methoxyxanthone (GJ-7), dihydr-okaempferol (GJ-8), naringenin (GJ-9), Kaempferol (GJ-10), quercetin (GJ-11), taxifolin (GJ-12),6-p-hydroxybenzyldihydroquercetin (GJ-13),8-p-hydroxybenzyl quercetin (GJ-14), dihydromorin (GJ-15), kaempferol-7-O-β-D-glucopyranoside (GJ-16), steppogenin (GJ-17), resveratrol (GJ-18), oxyresveratrol (GJ-19), butyro-spermol acetate (GJ-20), butyrospermol (GJ-21),(+)syringarenol (GJ22), umbelli-ferone (GJ-23),4-hydroxybenzyl ethyl ether (GJ-24),2,4-dihydroxybenzaldehyde (GJ-25),4-hydroxybenzaldehyde (GJ-26), vanillin (GJ-27),1,3,5-trimethoxy-benzene (GJ-28),2,4-dihydroxybenzoic acid ethyl ester (GJ-29),β-sitosterol (GJ-30),6-p-hydroxybenyl kaempferol(GJ-31),6-p-hydroxybenzyl quercetin (GJ-32), and (6S,12S,13S)-1-methoxycyanomaclurin (GJ-33). GJ-31-GJ-33were new compounds. GJ-1, GJ-2, GJ-14, GJ-23and GJ-25-GJ-29were isolated from this genus for the first time. GJ-3, GJ-13and GJ-32were isolated from this plant for the first time.The activity evaluation results showed that GJ-10and GJ-15had potent anti-H5N1virus activities with EC50at37.5and150μg/mL, respectively, and exhibited good dose-response relationships.
