The study is started from extract technology, screening of active parts, separation and identification of chemical componets; systematic research the quality of Cudrania tricuspidata (Carr.) Bur. through the aspect such as morphology, organ and chromatography technology, extractions and content determination, at the same time, institute the quality specification and draft illustration.(Ⅰ) Study on active parts chemical component: 1. Extract technology part: index on extraction rate, determine the best extract method is 70% alcohol, extracting 3 times, first 10 times alcohol 2 hours each, second, third 8 times alcohol 1 hour. 2. Through many kinds of chromatographic methods such as silica gel, and Sephadex LH-20, 9monomers have been isolated from the plant, including flavonoids, triterpene and steroidal. They are Stigmasterol(1), (13α,14β,17α,20R)-Lanosta -7,24-diene-3β-O-actate(2),β-sitosterol(3), Daucosterine(4), isocyclonorusin(5), Kaempferol(6), Taxifolin(7), 5,7,4'-trihydroxyldihydroisoflavone(8), Quercetin(9). Among them, monomers Stigmasterol(1), (13α,14β,17α,20R)-Lanosta -7,24-diene-3β-O-actate(2), isocyclonorusin(5), 5,7,4′-trihydroxyldihydro -isoflavone(8) are obtained from Cudrania tricuspidata for the first time. And monomers (13α,14β,17α,20R)-Lanosta -7,24-diene-3β-O-actate (2), isocyclonorusin(5), 5,7,4′-trihydroxyldihydroisoflavone(8) are obtained from the genius for the first time.(Ⅱ) Study on pharmacodynamics of anticancer: 1. Accroding to the screening experiment of vitro-anticancer active of Cudrania tricuspidata (Carr.) Bur., determine the active parts are the p. e. and chloroform parts. 2. Anticancer experiment of components shows that: Anticancer effects of Kaempferol and Taxifolin are fine; anticancer effects of Stigmasterol and (13α,14β,17α,20R)-Lanosta -7,24-diene-3β-O-actate are commen; anticancer effect of 5,7,4′-trihydroxyldihydroisoflavone is bad. So, use Kaempferol and Taxifolin as the reference substances of Cudrania tricuspidata (Carr.) Bur..(Ⅲ) Study on evaluation of species and quality: 1. Discrimination Cudrania tricuspidata and its adulterant Cudrania cochinchinensis, by the method of Herbais resrarch, resource, plants and character discrimption. 2. Study on discrimination of Cudrania tricuspidata (Carr.) Bur.: apply three method, they are microscopic, ultraviolet absorption spectra, thin layer chromatography, according to pharmacopeia of PRC. 3. Institute the limited standard in items of examination and extraction. 4. Determine the reference substances are Taxifolin and Kaemferol, determine the reference substances'content of Cudrania tricuspidata in different area by the method of HPLC, and institute the limited standard of reference substances.(Ⅳ) Protocol of quality specification: Institute the qualiy specification of Cudrania tricuspidata systimetic, including name, resource, macroscopic, discrimination (microscopic, physico-chemical), examination (moisture content, ash content and ash content which didn't resolve in acid), extraction, component determination, characters and meridian distribution, functions and indications, usage and dosage, storage and so on.
