| Objective This study was designed to explore the anti-anxiety and acute toxicity effectsof the Valeriana jatamansi valeportriates.Methods1.Under low temperature and drying circumstance smash the Valerianajatamansi root, and the powder of V. jatamansi was extracted exhaustively with95%aq.EtOH at room temperature3×24h, soaking time was24h before infiltration. Using4kinds ofvaleportriates content as an index, and four kinds of compound content has been determinedusing the HPLC.3. D101macroporous resin was used to separate and purify valeportriates.4.Using elevated plus maze test in rats, open field test and Light-dark transition test evaluatethe valeportriates anti-anxiety effect. And using the Elisa determines the corticosterone level.5. Using bliss method analysis the acute effect of V. Jatamansi extract.Results1.The petroleum ether fraction afforded5kinds of valeportriates compoundsand then selected relative high content of4kinds of compounds as the standard, whichcontain2kinds of new compound (Jatamanvaltrate Pand Jatamanvaltrate Q).2. HPLC testdisplaied the variation of regular pattern of4kinds of compound in Valeriana jatamansi. Andprovide the supplement proof of Valeriana jatamansi content determin in order to afford thecomplementary evidence of Valeriana jatamansi content control and reasonable utilize.3.The optimum purified process of D101macroporous resin was as follows: The washing dosage of30%ethanol was4BV to saturating the macroporous resin, the washing dosage of70%ethanol was8BV, and the washing dosage of85%ethanol was8BV, then85%ethanolportion was obtained, and it was further concentrated in vacuo to yield a residue and then useHPLC test the valeportriates content.4. Using classical experiment model such as light-darktransition test, open field test, elevated plus maze test and spontaneous activity test discussedthe anti-anxiety effect of Valeriana jatamansi valeportriates. These test result showvaleportriates have the anti-anxiety effect, and it was produced through triggered the HPAand regulated the neurotransmitters level. Test result show valeportriates reducedcorticosterone level and down-regulation NE, DA and5-HT level.5. The lethal dose50%(LD50) of crud extract and valeportriates are79.31g/kg and56.88g/kg, respectively. And itwere the793.1and568.8g/kg times for the usual dose of human body intake.Conclusion The valeportriates that extracted from Valeriana jatamansi has theanti-anxiety effect and acute toxicity result show it was relative security in clinical. So it haspotential role to become the anti-anxiety drug. |
