Objective: In primary Sj?gren’s syndrome(p SS),plasmacytoid dendritic cells(p DCs) are decreased in peripheral blood and accumμl ated in salivary glands, total glucosides of paeony(TGP)could attenuate DC maturation in vivo. We thus investigate the effect of TGP on p DCss in new p SS patients and its mechanism of anti-inflammatory effects.Methods: 17 new p SS without any treatment were recruited as disease group, accepted total glucosides of paeony for 3 months,17 healthy subjects as control group.The percentage of peripheral blood p DCsã€circulating plasma IFN-αã€IL-6ã€TNF-α levels were analysed by Flow cytometry and ELISA. Immunohistochemistry method has been used to observe p DCs infiltration in labial gland.Results: New p SS patients had significantly lesser p DCss in PB,and CXCR4 expressed by p DCs were higher than healthy controls.TGP could inhibited p DCs actiition and then reduced CXCR4 expression.Plasma IFN-α 〠IL-6 〠TNF-α levels of new p SS are increased, TGP reduce IL-6ã€TNF-α levels.Moreover, p DCs are present in labial gland of p SS under the effect of chemokines receptor CXCR4.Conclusion: Our findings reveal that activated pDCss transfer to the labial gland under the effect of chemokines receptor CXCR4.Total glucosides of paeony inhibited the activation of p DCss in primary Sj?gren’s syndrome. |
