| Objective:1 To explore the changes after Salvia miltiorrhiza Ligustrazine and glucose injection and Simvastatin in high-fat-fed rabbits.2 To investigate the effect of Salvia miltiorrhiza Ligustrazine and glucose injection combined with Simvastatin on the carotid restenosis after bollon injury in hyperlipidemia rabbits.Methods:Forty healthy rabbits(male or female) about 2.5-3.0kg were randomly divided into five groups, each group has 8 rabbits:1 Matched group(C), high fat diet for 10 weeks;2 Bollon injury group(H), high fat diet for 10 weeks;3 Simvastatin group(S), high fat diet while mixed with Simvastatin 2.2mg /(kg.d) for 10 weeks;4 Shenxiong glucose(SX) group, high fat diet while giving Salvia miltiorrhiza Ligustrazine and glucose injection intraperitoneal injection 5ml/(kg.d) for 10 weeks;5 Salvia miltiorrhiza Ligustrazine and glucose + simvastatin(SX + S) group, high fat diet which mixed with Simvastatin 2.2mg /(kg.d) and while giving Salvia miltiorrhiza Ligustrazine and glucose injection intraperitoneal injection 5ml /(kg.d) for 10 weeks.After eight weeks, H group,S group, SX group, SX + S group were received preoperative carotid intima balloon injury, C group was received sham surgery group. In the point of the test(0 weeks), 8 weeks and 10 weeks, we detected the rabbits’ weight, blood lipids, blood glucose levels(drawing blood from the rabbits fasting 12 hours). Respectively, the levels of TXB2, P-selectin by using ELISA were detected 24 h before and 3h,24 h,3d after operation. We detected the changes of pathology sections and morphology of rabbit carotid artery after two weeks.Results:1 Blood lipids and blood glucose levels: After 10 weekends,in H group, S group, SX group, SX + S group the rabbits’ TC, TG, LDL-C levels were increased(P <0.01). Compared with the group H,the TC, TG, LDL-C levels in S group,SX group and SX+S group were significantly lower(P <0.01), the difference was statistically significant(P <0.01). Compared with the S group, the TC, TG, LDL-C levels in SX + S group were decreased(P <0.05), HDL-C levels were increased(P <0.05). The difference of rabbit blood glucose levels has no statistically significant(P> 0.05).2 TXB2, P-selectin levels: The levels of TXB2 and P-selectin in H group were significantly higher than group C no matter before or after operation(P <0.01); after operation 3h, 24 h, 3d,we can see that the levels of TXB2 and P-selectin in group S, SX and SX + S were significantly lower than H group(P <0.01); compered with SX group, the TXB2, P-selectin levels in SX+S were significantly lower(P <0.05), and the SX + S group reduced more obvious.3 Restenosis: After two weeks, under the microscope,the rabbit carotid artery stained with HE has integrity structure with sommth endarterium in C group; H group showed intimal-thickening,intrabascular elastic plate boken,VSMC hyperplasia.There were a lot of foam cells and lipid deposition in intima and media; S group showed the degree of intimal hyperplasia was less than H group; compered with S group, SX group and SX + S group showed inhibitory effect is better, but the difference between SX group and SX+S group has no statistically significant(P> 0.05)..4 A comparison of all experimental groups damaged vascular intimal plaque area: compared with H group, S group, SX group, SX+S group significantly increased relative lumen area(P<0.01), intimal area, medial area and intimal / medial area decreased, significant difference(P<0.01); compared with SX group, S group, SX+S relative lumen area increased(P<0.01), intimal area, medial area and intimal / medial area were significantly reduced(P<0.01).Conclusion:1 Simvastatin can decrease the level of blood lipide.Simbastatin and Salvia miltiorrhiza Ligustrazine and glucose injection can weaken inflammatory reaction and microthrombosis in rabbits aftere carotid balloon injury;2 Salvia miltiorrhiza Ligustrazine and glucose injection combining with simvastatin can significantly reduce the reaction of inflammation and platelet activation,which has more effection than simvastatin;3 Our study indicate that compared with the group whithout simvastatin,simbastatin group shows more significantly effection on weaken thereaction of inflammation and platelet activation.These results confirm that both simvatation and Salvia miltiorrhiza Ligustrazine and glucose injection can control intimal hyperplasia and prevention of vascular restenosis.
