Objective:By studying the influences of musical electro-acupuncture on the activityof substantial nigra Ca2+-Mg2+-ATPase and the dopamine neuronsapoptosis in the brains of the Parkinson’s disease (PD) model rats toinvestigate the injured neurons protection mechanism in the treatment ofPD.Methods:1. The rotation model of PD rats was established by injecting6-hydroxydopamine (6OHDA) into the double targets in right striatum.2. SD rats were randomly divided into5groups: the electro-acupuncturegroup, Madopar group, model group, sham-operation group and blankcontrol group. For subjects in the electro-acupuncture group, penetrationacupuncture was used in the temple along the Baihui acupoint, and thenconnected with the electro-acupuncture. For those in Madopar group,intragastric administration of levodopa and benserazide tablets was givento them.3.After28days, the rats in each group were sampled underanesthesia respectively. Their brain tissue was fixed through perfusionwith4%paraformaldehyde(PA), and the substantia nigra was separatedand saved. Coomassie Brilliant Blue method was used to measure theprotein of the tissue. Activity of nigra Ca2+-Mg2+-ATPase wasdetermined by spectrophotometry. Flow cytometry was adopted to detectthe dopamine neurons apoptosis rate in rats’substantia nigra. In situ endlabeling method was used to detect apoptosis. SPSS18.0was used forstatistical analysis of the obtained data.Results:Ca2+-Mg2+-ATPase activity of substantial nigra are higher in musicalelectro-acupuncture group and Madopar group than that of the modelgroup, with a very significant difference (P<0.01); levels of apoptosis indopamine neurons of the substantia nigra are lower in the musical electro-acupuncture group and Madopar group than that of the modelgroup, and the difference is significant (P<0.05); compared with theblank control group, indexes in the model group are obviously increased(P<0.01); but the differences between musical electro-acupuncture groupand Madopar group is not significant (P>0.05); and there is also noobvious difference between the blank control group and thesham-operation group (P>0.05); there is no significant difference betweenthe sham-operation group and Madopar group as well as musicalelectro-acupuncture group (P>0.05).Conclusion:1. Musical electro-acupuncture has therapeutic effect on PD model rats;2.Musical electro-acupuncture therapy can up-regulate the abnormallydecreased Ca2+-Mg2+-ATPase activity;3. Musical electro-acupunctureplays a role in protecting the dopamine neurons in the substantial nigra ofthe PD model rats. |
