| Objective Spectrophotometric method is used to test the effect ofdifferent electroacupuncture scalps point-through-point therapy on theactivity of substantia nigra including Na~+-K~+-ATPase in the brain of ratwith parkinson’s disease and further discuss the action mechanism oftreatment of different electroacupuncture scalps point-through-point therapyfor PD.Methods double targets microinjection of6-hydroxy dopamine in rightcaudate putamen was used to prepare hemi-parkinson rat rotational model;musicoelectroacupuncture and impulse electroacupuncture scalpspoint-through-point therapy were as the treatment method; the model groupwas taken as the control group and black group, sham operation group,madopar group, electroacupuncture group and impulse electroacupuncturegroup were also set.(1) After the model rats with PD when the period ofobservation was ended were treated with anesthesia, method of brain tissueperfusion fixation with4%paraformaldehyde was used to separate and storethe substantia nigra.(2) Coomassie brilliant blue method was used to test theprotein content.(3) Spectrophotometric method was used to detect the activityof substantia nigraNa~+-K~+-ATPase in the brain.(4) SPSS17.0software wasused to analyze the obtained data and calculate the mean value, standarddeviation. Test and analysis of variance were used for analysis ofmeasurement data.Results The substantia nigra Na~+-K~+-ATPase activity in the brain of ratof musicoelectroacupuncture group, impulse electroacupuncture group andmadopar group was significantly higher than that of model group, withstatistically significant difference(P<0.01); the activity of model group wassignificantly lower than that of normal group and sham operation group; thedifference of activity between musicoelectroacupuncture group and impulseelectroacupuncture group as well as madopar group was not statistically significant(P>0.05); madopar group and impulse electroacupuncture grouphad statistically significant(P<0.05); there was no statistically significantdifference between black group and sham operation group(P>0.05).Conclusions1Activity of substantia nigra Na~+-K~+-ATPase in thebrain of rat with PD is significantly increased in therapy group.2Themusicoelectroacupuncture scalps point-through-point therapy group is moresignificant than that of impulse electroacupuncture group.3From the angle ofcell and molecular biology,it further revealed mechanism of action of PDwith musicoelectroacupuncture scalps point-through-point therapy andimpulse electroacupuncture scalps point-through-point therapy. |
