Controlled-Release Oxycodone Alone Or Combined With Pregabalin For The Management Of Malignant Neuropathic Pain

Objective:Observation of oxycodone hydrochloride controlled-release tablets combined pregabalin tre atment MNP clinical effects and adverse reactions, specifically oxycodone hydrochloride controlled-r elease tablets combined use of pregabalin alone whether oxycodone hydrochloride controlled-release tablets have the same analgesic effect.Methods:Choose the cancer patients in pain clinic, including moderate pain patient and severe pain patient (NRS>4). Aging40～80,expected to live for30days or more,taking orally Oxycodone for a week,Initial dose is10mg, Once every12hours,According to the situation of pain relief to adjust. On the eighth day patients (NRS<4) into A group,Continue to Oxycodone treatment for2weeks. NRS>4into B group, B divided into B1and B2groups. B1group continue to take orally Oxycodone for2weeks.According to the change of pain increasing and decreasing the dose. B2group keep the eighth day’s dose, joined the Pregabalin relieve the pain.The initial dose of Pregabalin is75mg,2times a day. Record the dose and side effect and the scores of life quality.Results:81patients finished the experiment. A group28cases, B1group27cases, B2group26cases.1. The contrast of the degree of pain relief:In the first stage of experiment, the pain relieved obviously, From the first day to the eighth day the pain score decreased obviously; in the second stage,the pain score of the three groups of patients reduced compared the fifteenth day to the eighth day, there is no difference between the22th day and the15th day. The pain score reduced in compared B1group to B2group in the22th day, but no statistical significance.2. The contrast of the dose between Oxycodone and Pregabalin:the dose of Oxycodone increased in A, B1group in the8th,15th,22th day, the dose of Oxycodone keep the same, but the Pregabalin increasd in B2group after united Pregabalin in the8th day. we also find that the dose of Oxycodone in B2group reduced compared with B1group. There is statistical significance(P<0.05).3. The contrast of the the incidence of adverse reactions:Major adverse reactions in treatment process is nausea, vomiting, constipation, dry mouth, drowsiness, dizziness, skin pruritus, etc. The adverse reactions reduced or disappeared after symptomatic treatment. but there is no statistical significance in B1ang B2.4. The contrast of the quality of life:the quality of life scores declined compared the22th day to lth day in3groups, the treatment is proved effective, can improve the quality of life of patients, but there is no statistical significance in the score of the quality of life in B1and B2.Conclusions:Oxycodone hydrochloride controlled-release tablets treatment of neuropathic pain in cancer poor results, you can choose a joint pregabalin treatment, combination therapy can reduce the dosage of oxycodone.
