Objective:To detect the changes in the distribution of T cell subsets (CD3+CD4、CD3+CD8and Treg) from healthy female adult peripheral blood after co-cultured with the culture supernatant AO-I concentrations induced cells; Explore whether AO-I can down-regulate MyD88+human ovarian cancer cells-mediated immunosuppression.Methods:Different concentrations of AO-I induced MyD88+human ovarian cancer cells, get the culture supernatant,co-culture with the T cell Separated from healthy female adult peripheral blood, Flow cytometry instrument was used to detect the percentage of T lymphycote subsets (CD3+CD4+、CD3+CD8+and Treg), and calculated the ratio of CD4+/CD8+.Results:1、The percentage of CD3+CD4+in SKOV-3culture supernatant disposed groups (40.453±0.270) was lower than the normal T cell groups (48.247±0.385), the percentage of CD3+CD8+(30.233±0.258) was higher than it (26.580±0.295). The percentage ratio CD4+/CD8+(1.338±0.016) was Significantly lower than normal T cell groups (1.815±0.029),(P<0.05); The percentage of Foxp3in SKOV-3culture supernatant disposed groups (10.260±0.782) was Significantly higher than normal T cell groups (7.340±0.458),(P<0.05);2、The percentage ratio of CD4+/CD8+in SKOV-3culture supernatant disposed groups which induced by different AO-I concentrations groups,100umol/L concen-tration groups(1.742±0.063),50umol/L concentration groups(1.434±0.032) and10umol/L concentration groups(1.358±0.018)were higher than the SKOV-3culture supernatant disposed groups(1.338±0.016),the higher concentration of AO-I are, the ratio higher (P<0.05); The percentage of Foxp3in SKOV-3culture supernatant disposed groups which induced by different AO-I concentrations groups,100umol/L concentration groups(2.667±0.180),50umol/L concentration groups(3.547±0.309)and lOumol/L concentration groups(4.573±0.191) were lower than it (10.260±0.782), the higher concentration of AO-I are, the percentage lower (P<0.05);3、The percentage of CD3+CD8+in SKOV-3culture supernatant disposed groups which induced by LPS (33.573±0.828)was higher than the SKOV-3culture super-natant disposed groups(30.233±0.258);The percentage of CD3+CD4+(29.740±0.675) was lower than it(40.453±0.270); The ratio CD4+/CD8+(0.890±0.028) was also lower than it(1.338±0.016),(P<0.05);The percentage of Foxp3(10.807±0.430) was higher than it (10.260±0.782),(P<0.05);Conclusions:1、KOV-3cells can autocrine a lot of immunosuppressive factors, the culture supernatant can inhibit CD3+CD4+T cells expression, increase CD3+CD8+T cells expression relatively, reduce the CD4+/CD8+ratio, promote Treg expression, and make the body in a state of immunosuppression.LPS can promote it.2、AO-I can cut MyD88+SKOV-3people ovarian cancer cells secrete immune suppressors by blocking TLR4/MyD88pathway, in a certain concentration range and dependent manner, thus promote CD3+CD4+T cells expression, decrease CD3+CD8+expression,increase CD4+/CD8+ratio, stimulate the body’s immune function; Inhibi-ting Treg expression, reduce the distribution proportion, ease the body’s immune inhibition, down-regulate MyD88+human ovarian cancer cells mediated immune inhibition.3、AO-I can down-regulate MyD88+human ovarian cancer cells mediated immune inhibition by blocking TLR4/MyD88pathway, can strengthen immunity, antitumor, and other functions, is expected to be an effective new drug, in clinical immune-therapy for ovarian cancer.
