The Experimental Study On The Effect Of Qi Lian Fu Zheng Capsule Combined With Intraperitoneal Perfusion In Mice Model Of H22 Hepatoma Cell

Objective:Observe the effect of different treatment groups by establishing models of mice with H22 hepatoma cell and giving the treatment of QiLian FuZheng capsule gavage combined with intraperitoneal perfusion chemotherapy, and provide a theoretical basis for the clinical application of QiLian FuZheng capsule combined with 5-Fu chemotherapy.Methods:Through establishing models of mice with liver cancer by injecting H22 hepatoma cell in the liver and through intervening in mice with different drug groups, observe the general situation, anatomy and pathology of mice and test the expression level of HGF、c-Met gene by RT-PCR and the expression level of HGF、c-Met protein by Western Blot.Results:1、The result that weight change of mice with different drug groups before and after treatment shows QiLian FuZheng capsule gavage combined with intraperitoneal perfusion can improve the general situation of mice and increase the weight of mice significantly(P<0.01). 2、The result that inhibition rate of tumor of mice with different drug groups shows QiLian FuZheng capsule gavage combined with intraperitoneal perfusion has significant effects of antitumor(P<0.01). 3、Test the expression level of HGF、c-Met mRNA of hepatoma cells of mice with different drug groups by RT-PCR and the result shows QiLian FuZheng capsule gavage combined with intraperitoneal perfusion can lower the expression level of HGF、c-Met mRNA significantly(P<0.01).4、Test the expression level of HGF、c-Met protein of hepatoma cells of mice with different drug groups by Western Blot and the result shows QiLian FuZheng capsule gavage combined with intraperitoneal perfusion can lower the expression level of HGF 、 c-Met protein significantly(P<0.01).Conclusions : The treatment of QiLian FuZheng capsule combined with 5-Fu chemotherapy has significant effects of antitumor, and the downregulation of expression level of HGF、c-Met gene may be one of the mechanisms of inhibiting the growth of tumor.
