Objective To investigate the influence of polymorphism and Gene-Gene Interaction among 5-HTTLPR、5-HT2 A receptor polymorphism and DβH-1021C/T polymorphism on attention deficit hyperactivity disorder in children. Methods Polymerasechain reaction(PCR) and RFLP technique were used to test 5-HTTLPR,5-HT2 A receptor T102 C polymorphism、DβH-1021C/T polymorphism. We used χ2 test to analyze the infiuence of polymorphism on ADHD. We used t test to analyze the differences of polymorphism on the behavior prombles of ADHD.We used logistic regression to analyze the Gene- Gene Interaction. Results There was difference in the frequency of 5-HT2 A receptor T102 C polymorphism between ADHD group and controls(χ2=5.28,P=0.02),and there wasn’t difference in the frequency of 5-HTTLPR、DβH-1021C/T polymorphism between ADHD group and controls(χ2=0.24,P=0.62; χ2=0.01,P=0.93); The interaction of 5-HTR2 A T102C and 5-HTTLPR maybe have the trend of in children with ADHD(OR=2.00,P>0.05), the interaction of 5-HTR2 A T102C and DβH-1021C/T maybe have the trend of in children with ADHD(OR=2.80,P>0.05), the genotype of 5-HTTLPR、5-HTR2 A T102C and DβH-1021C/T haven’t the interaction in children with ADHD.There were differences in impulse hyperactivity scores and anxiety scores about SS/SL genotype and LL genotype of 5-HTTLPR(P=0.007, P=0.010), the scores of impulse hyperactivity in LL genotype is highter thanSS/SL genotype, the scores of anxiety in SS/SL genotype is highter than LL genotype;There were differences in conduct promble scores, anxiety scores and hyperactivity index scores about TT genotype and TC/CC genotype of 5-HTR2A(P=0.040,P=0.019,P=0.038),the scores of conduct promble, anxiety, hyperactivity index in TC/CC genotype is highter than TT genotype;There weren’t differences in behavior prombles about CC and CT/TT of DβH-1021C/T.Conclusion 5-HTR2 A T102C polymorphism have association of ADHD; The genotype of 5-HTTLPR,5-HTR2 A T102C and DβH-1021C/T haven’t the interaction in children with ADHD, The scores of conduct promble, anxiety, hyperactivity index in TC/CC genotype of 5-HTR2 A is highter than TT genotype of 5-HTR2 A. |
