Euphorbia sororia seed is part of the fruit on the ground, a kind of medicinal plants in Xinjiang, its belong to Euphorbiaceae Euphorbia, there are many in Hotan, This herbal has been used in Uighur medicine for a long time, Uighur medicine commonly used the seeds to cure various diseases. Euphorbia sororia and its seeds have been as the main component in the use of some prescription medicine. The chemical composition of the seed has many pharmacological activities, such as good effective for the brain, stomach and eyes, antibacterial, anti-HIV virus, cell toxicity. Other ingredients also used to treat for the treatment of stomach pressure, chest pain, skin disease, stroke, appetizers, and other ailments. 6,7-Dihydroxycoumarin and its derivatives have a variety of pharmacological effects with antibacterial, anticancer, antioxidant. Therefore, it is of great significance to study the synthesis of 6,7-dihydroxy coumarin and its derivatives. The contents of this paper include the purification of the seeds of Euphorbia sororia, antioxidant activity, chemical composition and the synthesis of 6,7-dihydroxycoumarin derivatives.1. n-Butanol extract of Euphorbia sororia seed was purified using AB-8 macroporous resin. In order to determine the optimum purification conditions for total flavonids and coumarin, the sample volume, desorption, sample concentration, adsorption flow rate, eluent concentration, eluent dosage and inquiry were investigated: the result indicated that the sample volume 10 mL/g, desorption 80 mL,the sample concentration 0.04 mg/mL, adsorption velocity control 1 mL /min, eluent concentration 70% ethanol solution, eluent 90 mL have to provide best result. The purification method can purify total flavonoids and of coumarin of Euphorbia sororia seed effictively by AB-8 macroporous adsorption resin.2. The research of petroleum ether layer of Euphorbia sororia seed were measured with vitamin C control, to eliminate ·OH and DPPH· free radical and reducing power, the experimental results show that petroleum ether layer of the seeds of Euphorbia sororia has a strong ability of scavenging hydroxyl radical, and DPPH radical scavenging rate is lower than the vitamin C, but still has a certain reduction capacity, stronger antioxidant ability in vitro.3. Extract part of Euphorbia sororia seed were separated by column chromatography. As a result, six compounds were isolated, and its structure were comfirmed by IR, 1H NMR, 13 C NMR chromatographic method. The compounds include of 6,7-dihydroxycoumarin, acacetin, acacetin-7-O-β-D-glucopyranoside, Sororianolide B, β-Amyrin-3-O-β-D-glucopyranoside, β-sitoterol.4. 6,7-Dihydroxycoumarin isolated from Euphorbia sororia seed as a starting materials, which reacted with 2-bromoethanol, ethyl 2,3-dibromopropionate, 3-bromopropyne, 1,2-dimethylbenzene in toluene respectively, under different catalyst, to offord a series of 6,7- dihydroxycoumarin derivatives.
