Bear bile as ursidae animals bear(Selenarctos thibetanus Cuvier.) or brown bear(Ursus arctos L.) dry gallbladder. In pharmaceutical preparations, wild bear bile had been replaced by drainage bear bile powder, from a certain extent, the contradiction between the clinical medication and the protection of animals was be eased, but the fundamental problem did not solved, substitute for bear bile has became a problem to be solved. Rabbit bile compared with bear bile had similar sexual flavour, chemical components and pharmacological effects, and rabbit bile powder was easy obtained, had stable sources and low price, all of them were provided rabbit bile powder substitute for bear bile.Rabbit bile powder and bear bile powder were compared about the similarities and differences in the pharmacognosy identification. And two kinds of powder were compared by using thin layer chromatography, and for further research, two kinds of powder were compared by using LC- MS and FT-IR method. The contents of total bile acids of two kinds of powder were determined by sulfuric acid- furfural reaction, and the amino acids contents of two kinds of powder were determined by automatic amino acid analyzer, and the element contents of two kinds of powder were determined by inductively coupled plasma-atomic emission spectrometry method. This paper used the ammonia lead mice cough and phenol red secretion evaluating rabbit bile powder and bear bile powder cough expectorant effect, and two kinds of powder analgesic action were evaluated by the method of hot plate and acetic acid twisting tests, the anti-inflammatory action of two kinds of powder were investigated by using the mice ear swelling induced by xylene method.The experimental results as follows: Rabbit bile powder and bear bile powder were identificated by using microstructure, physical and chemical, thin layer chromatography and thin layer chromatography, it found that both kinds of bile powder have certain differences in composition; Two kinds of powder were analyzed by the method of LC-MS and FT-IR, it found that oxygen cholic acid was existed in rabbit bile powder, same as to bile acid, chenode oxycholic acid; Ursodeoxycholic acid, chenodeoxycholic acid and cholic acid were found in bear bile powder; Total bile acid contents in the two kinds of powder had been determined by sulfuric acid- furfural reaction, total bile acid contents were metered with cholic acid, total bile acid contents in rabbit bile powder and bear bile powder respectively were 23.33%, 16.45%, total bile acid contents were metered with ursodeoxycholic acid, total bile acid contents in the two kinds of powder were 10.03%, 25.51%. Amino acid determination results showed that there were differences between the two kinds of powder on types of the amino acids, only tryptophan in rabbit bile powder, but three such as tryptophan, histidine, isoleucine in bear bile powder not be checked out. In the detection of amino acids, except aspartic acid, glutamic acid, valine, leucine, tyrosine were more in bear bile powder, the rest of the amino acids contents were more in rabbit bile powder; The results showed that the contents of trace elements such as Ca, Mg, Mn, Fe, K,Zn were higher in rabbit bile powder than in bear bile powder, Na and Cu contents were higher in bear bile powder than rabbit bile powder, both of two kinds of powder the heavy metal elements Cd and Pb were not be detected. Antitussive expectorant results showed that both powder had a certain extent antitussive effect,both of them can obviously increased the cough incubation period, decreased the times of cough, and there existed a certain positive correlation between antitussive effects and doses; Two kinds of powder expectorant effect had a good, similar pharmacological effects.Analgesic action results showed that two kinds of powder had a certain analgesia effect, they can increased the pain threshold and reduced the number of twist body, anti-inflammatory effects results showed that two kinds of powder had a certain anti-inflammatory effects, and rabbit bile powder anti-inflammatory effect better than bear bile powder.In this paper, the following conclusions had be got: By comparing pharmacognosy, some differences between two kinds of bile powder had be found, and combined with the determination of the chromatography and spectral methods, rabbit bile powder and bear bile powder contents had some in common and some in different, and rabbit bile powder contains substances have be found relatively more in bear bile powder, which should be further analysis; There had no obvious differences in amino acids types and quantities of two kinds of bile powder, the amounts and types of amino acids of rabbit bile powder a bit more than bear bile powder; No Cd, Pd or other toxic elements were been determined in both kinds of powder. Pharmacological experimental results showed that both kinds of powder had antitussive, expectorant, analgesic and anti-inflammatory effects, there existed a certain positive correlation between antitussive effects and doses; In terms of antitussive, expectorant and analgesic, two kinds of powder had a similar pharmacological effects,and had no obvious differences; And anti-inflammatory effects, rabbit bile powder effect was better.
