Objective To dtect the relative quantitative measure of Musashi-1 and β-catenin in tissue of shed and unshed endometrium in first day of menstruation, investigate the ultrastructure of endometrium at day 1 and 5 of the cycle of endometriosis. In ordor to explore the roles of ultrastructure changes and molecular biology in endometriosis.Methods Collect shed and unshed endometrium in first day of menstruation. 36 tissues from patients with endometriosis for experimental group, 40 tissues from patients with benign ovarian tumor and infertility result from fallopian tubes disease for control group. Randomly selecte ectopic endometrium at day 5 of the cycle with or without endometriosis, totally eight cases. SYBR Green fluorescence probe Real Time-PCR to test the relative quantitative measure of Msi-1 and β-catenin in tissue of shed and unshed endometrium in first day of menstruation with or without endometriosis.To compare the ultrastructure of shed and unshed endometrium in first day of menstruation and the ectopic endometrium at day 5 of the cycle with or without endometriosis by transmission electron microscope. Results 1.The expression of Msi-1 in unshed endometrium is higher 1.19 times than that in shed endometrium in experimental group,the difference has no statistical significance(P >0.05), while that’s higher 11.31,3.66 times than that in shed and unshed endometrium of control group, has statistical significance(P < 0.05).2. The expression of Msi-1 in shed endometrium of experimental groupis higher 9.51,3.07 times than that in shed and unshed endometrium of control group, has statistical significance(P < 0.05).3. The expression of β-catenin in unshed endometrium is higher 1.53 times than that in shed endometrium in experimental group,and higher 164.28,4.92 times than that in shed and unshed endometrium of control group, has statistical significance(P < 0.05).4. The expression of β-catenin in shed endometrium of experimental groupis is higher 107.63, 3.23 times than that in shed and unshed endometrium of control group, has statistical significance(P< 0.05). 5. Among shed and unshed endometrium in first day of menstruation of endometriosis,the expression of Musashi-1 and β-catenin has positive correlation(r=0.571,P=0.05 r=0.485,P=0.041),whiel there are no coreelation in shed and unshed endometrium in first day of menstruation of control group,(r=-1.31,P=0.582, r=0.431,P=0.058). 6.Compare to the ultrastructure of control group: gland secretion of epithelial are not of uniform size and disorganized;the nucleus variation are obviously; mitochondria, ribosomes and other organelles rich in the unshed endometrium in first day of menstruation of patients with endometriosis.The rough endoplasmic reticulum is well developed;the volum of some mitochondrial increases;and organelles rich in the shed endometrium in first day of menstruation of endometriosis. The cilia and microvilli of secretion cell are abnormal increase and elongate; many nuclear atypia, intracytoplasmic mitochondria, rough endoplasmic reticulum and other organelles are abnormal rich in ectopic endometrium at day 5 of the cycle of endometriosis. Conclusion 1.The stem cell maker Musashi-1 may through regulate the Wnt/β-catenin signal path to contribute the formation of endometriosis.2. The endometrium ultrastructure changes result in the changes of function, the activated endometrium can develope into endometriosis with other appropriate conditions finally.
